Streszczenie angielskie: Thioredoxin reductase (TrxR) is the first selenoenzyme containing selenocysteine in the active center and FAD as a second prosthetic group. TrxR catalyses the NADPH-dependent reduction of thioredoxin an of many other physiologically important substrates. TrxR exhibits a many-fold increase in the activity in tumor cells and stimulates their proliferation as well the phenotype changes. Some gold compounds and a number of other clinically and experimentally tested drugs have been shown to inhibit TrxR. The involvement of TrxR/Trx/NADPH system in a broad spectrum of cellular processes renders it a potential target for therapeutic approaches.

    Streszczenie angielskie: A systematic investigation into the impact of the substituents introduced into the indolo[2,3-b]quinoline system is described. The findings clearly demonstarte that the compounds bearing a methyl group or a longer aliphatic chain at the N-6 position are inactive against prokaryotic and eukaryotic cells. The indroduction of alkyl-amino-alkyl substituent at the N-6 position of indolo[2,3-b]quinoline accounts for the appearance of the antimicrobial and cytotoxic properties. The cytotoxicity against oral epidermoid carcinoma KB(ID50) is in the range from 2.0 to 9.0 ćM, and the antimicrobial activity (MIC) falls between 0.003 and 0.50 mM. The structural relation within 6H-indolo[2,3-b]quinolines, concering their antimicrobial and cytotoxic activity, corresponds well with their ability to bind DNA and to inhibit topoisomerase II activity.

    Streszczenie angielskie: A hypotehsis of existing cause-effect relationship between the disturbances of antioxidative balance, as a biomarker of occupational exposition to nitrogen oxides and ozone, concentration of selected trace elements in blood serum and hair, taken as biomarkers of exposition, and impairment of the respiratory system, regarded as a biomarker of effect has been tested on a representative goup of welders occupationally exposed to these pollutants. The subjects of the study were 94 welders aged 41.2 (+10.0) exposed in their workshops to nitric oxides and ozone in concentration exceeding TLV. As a control group served 115 workers, 40.8 (++10.2) years old, performing virtually the same tasks, except welding. It was found that in the group of welders concentrations of TAS, CT, SOD were lower compared to the controls. COPD and Small Airway Disease (SAD) in the welders groups was more frequent than in the controls (COPD:14.9 p.c. welders, 3.5 p.c controls; at risk of COPD 37.2 p.c. welders, 15.7 p.c controls; SAD:32 p.c. and 20 p.c respectively). From a wide array of trace elements in blood and their taken into account in principal components analysis (PCA), and mutual contributions analysis (MCA), only the relation of MEF50, Cu in blood serum and TAS were statistically valid. MCA revealed that asymmetrical interactions exist between TAS and Cu (ëCu-TAS = 97.2 p.c., ëTAS-Cu = 36.1), as well as between TAS and MEF50 (ëMEF50-TAS = 82.4 p.c., ëTAS-MEF50 = 23.9). The Concentration of copper in blood serum correlacted better with antioxidant status (TAS) than the content of this element in hair.
