    Streszczenie angielskie: Stability of gene expression and transffection efficiency plays the main role in the application of gene transfere method. In somatic cell gene delivery, expression of the gene product is limite by the function of the cell to which it is delivered. In te present study analyzing the lipofected adherent cells, we have shown that lower level of transgene: á-galctosidase activite at later time period correlated with decrease in cell viability, which was shown to be due to apoptosis. Apoptosis following DNA uptake occurred only when DNA was present during lipofection.

    Streszczenie angielskie: The long QT syndrome (LQTS) is a disorder of ventricular repolarization that exposes affected individuals to cardiac arrythmias and sudden death. The first gene for LQTS has been mapped to chromosome 11 p. 15.5 by genome-wide linkage analysis. This gene, originally named KVLQT1 (and later KCNQ1), is a novel potassium channel gene. Mutations in the human KVLQT1 gene, encoding the ŕ-subunit of the KVLQT1 channel, cause the long QT syndrome. In this work, we analysed the sequence of six KVLQT1 exons in patients with various heart pathologies. We describe 6 different mSSCP patterns with no disease-related SSCP conformes in any sample. Different sequencing of exons 2 to 7 confirmed the absence of mutations. This suggests that the analysed region of the KVLQT1 gene is not commonly involved in pathogenesis of the long QT syndrome.

    Streszczenie angielskie: Gut-derived adenocarcinoa Caco-2 cells were treated with sodium butyrate (NaB) at physiologically relevant concentrations. We characterized its effects on proliferation, differentiation, apoptosis, adhesion to the solid support and interleukin-8 secretion. Differentiation was determined by brush border alkaline phosphoatase activity. Apoptosis was assessed by acridine orane and Hoechst stains. Differentiation and apoptosis were analyzed in both adherent and floating cell populations. The transformed Caco-2 cells did not retain their malignant phenotype in the presence of NaB. They appeared to undergo a change in the phenotype induced by NaB, as indicated by reduced proliferation, enhanced differentiation, stimulation of apoptosis leading to decreased viablility of cells, and stimulaiton of interleukin-8-secretion. Considering all the above facts and data, we postulate that Caco-2 cells cultured in NaB supplemented medium could regain the phenotypic characteirstics of the phenotype of the parent cell from which originated the caco-2 line.

    Streszczenie angielskie: Background: Although HCV and HBV are essentially hepatotropic, several lines of evidence suggest that these viruses can infect other cells, also PBMC in most patients with chronic HCV. Material/Methods: The presence of HBV DNA and HCV-RNA was determined by a polymerase chain reaction (multiplex PCR and RT-PCR -nested PCR) in a group of patients with chronic liver disease. HCV-RNA was investigated in serum, plasma and peripheral blood mononculear cells (PBMC) while HBV-DNA only in serum or plasma. Results: Among 374 patients tested, HCB-RNA was detected in the venous blood of 208 patients; HCV RNA alone was detected in 154 patients and 54 patientwere co-infected by HCV and HBV. HBV-DNA was found in 128 of 374 patients, while infection by HBV only was found in 74 patients. It was also shown that in the presence of HBV the replication ability of HCV is lower (p = 0.085, Goodman-Kruskal Gamma = 0.561 and YuleQ = 0.5610). Conclusions: Since coexistence of HBV adn HCV is not a rare cases, diagnostics of hepatitis cannot be limited to detection of one type of the virus only. Nisinterpretation of the virus type that caused the infection may lead to serious complications, especially in those cases when interferone is used for treatment.

    Streszczenie polskie: W pracy przedstawiono znaczenia programowanej śmierci komórki (apoptozy) w patogenezie schorzeń układu naczyniowego na przykładzie tętniaka aorty. Omówiono charakterystyczne cechy zjawiska apoptycznej śmierci komórki, uwzględniając najczęstsze bodźce stymulujące oraz czyniki regulacyjne - szczególnie białka rodziny Bcl-2 oraz receoptory śmierci komórki. Na podstawie współczesnej, wieloczynnikowej teorii powstawania tętniaka przedstawiono znaczenie opoptycznej śmierci komórki w zmniejszeniu liczby komórek błony środkowej aorty, charakterystycznym dla tętniaka aorty oraz torbielowatego zwyrodnienia błony środkowej. zwrócono uwage na znaczenie komórek zapalnych (limfocyty, makrofagi) oraz możliwe czynniki wywołujące apoptyczną śmierć komórki u chorych ze zmianami w obrębie błony środkowej aorty.

    Streszczenie angielskie: In the paper a role of programmed cell death (apoptosis) in the vascular disease development was presented. The influence of apoptosis on the aortic aneurysm development was discussed. Characteristic cell changes concerning apoptotic cell death were described, including inducers and regulatory factors - especially Bcl-2 protein family and death receptors. According to the current multivariant theory concerning aneurysm etiology, apoptosis can play the role in the decrease of the smooth muscle cell number in the medial aortic layer in the cases of aortic aneurysm and cystic medial degeneration. The role of inflammatory cells such as lymphocytes and macrophages and the factors inducing apoptotic cell death in the degenerative changes of the aortic wall is emphasized.

    Streszczenie angielskie: Subjects: P53 overexpression or mutations correlate with poor prognosis and high prolifeartive activity in human tumors. Aim. The aim of the study was to evaluate the usefulness of P53 and histone 2Hb/H4 genes expression level as a marker of vulvar carcinoma. Material and methods: The level of P53, 2Hb and H4 mRNA was estimated using RT-QPCR method in vilvar carcinoma (n = 27) at vartious stages of advancement of the neoplastic process. Results: In our experiments the average H2b and H4 genes expression was significantly higher lymph nodes in comparison of normal lymph nodes. Ca vulvae G2 showed significantly higher P53, H2b and H4 mRNA levels than normalk tissues. Ca vulvae G3 showed higher P53 and H2b mRNA levels and lower H4 mRNA level than normal tissues. Conclusion: The results confirm advantage of estimation of P53, 2Hb and mRNA levels as a marker of vulvar carcinoma. Overexpression of H2b encoding gene appears to be connected with genetic instability.

    Streszczenie angielskie: KCNQ1 (formerly called KVLQT1) is a Shaker-like voltage-gated potassium channel gene responsible for teh LQT1 sub-type of LQTS. In general, heterozygous mutations in KCNQ1 cause Romano-Ward syndrome (LQT1 only), while homozygous mutations cause JLNS (LQT1 and deafness). To date, more than 100 families with mutations in this gene have been reported, most with their own novel 'private' mutations. The majority of these mutations are missense. However, other types of mutations, such as deletions, frame-shifts and splice-donor errors have also been reported. There is one frequently reported mutated region (the 'hotspot'). KCNQ1 is now believed to be the most commonly mutated gene in LQTS. The combination of normal and mutan;t KCNQ1 ŕ-subunits has been found to form abnormal I KS channels, hence mutations associated with the KCNQ1 gene are also believed to act mainly through a dominant-negative mechanism (the mutatnt form interferes with the function of the normal wild-type form through a 'poison pill' type mechanism) or loss of function mechanism. Even in the case of carriers of teh same mutaion, it is currently unknown why there are significant clinical phenotype variations in LQT1 patients. This question could be answered by increasing the number of patient genotypes studied. LQT1 patients expereince a majority of their cardiac events (62 p.c.) during exercise, and only 3 p.c. occur during rest or sleep. Of the patients who experienced cardiac events while swimming, 99 p.c. were LQT1. Auditory stimuli are rare and occur in only 2 p.c. of patients. However, both lethal and non-lethal events followthe same pattern.

    Streszczenie polskie: Poza funkcją bariery, komórki nabłonkowej jelita (IECs) odgrywają miejscową rolę immunologiczną. Nabłonek jelitowy uczestniczy zarówno we wrodzonej, jak i nabytej odpowiedzi immunologicznej. Bezpośredni udział IECs w odporności wrodzonej realizuje się poprzez sekrecję wielu substancji, które przyczyniają się do tworzenia czynnościowej bariery śluzówkowej. IECs odgrywają również ważną rolę w nabytej odpowiedzi immunologicznej poprzez zdolność regulowania miejscowych funkcji immunologicznych limfocytów śródbłonkowych (IELs) oraz tkanki limfoidalnej zlokalizowanej pod warstwą nabłonkową. IECs stanowią pierwszy potencjalny typ komórek prezentujących antygen w kontakcie z obcymi antygenami obecnymi w świetle jelita. Podczas infekcji mediatorów zapalnych IECs zwiększją ekspresję cytokin i cząsteczek adhezyjnych. Cytokiny stymulują napływ i aktywację komórek immunologicznych/zapalnych, natomiast cząsteczki adhezyjne odpowiadają za ich adhezję w miejscu infekcji. Dwukierunkowa interakcja pomiędzy IECs i IELs odgrywa również ważną rolę w lokalzacji limfocytów w warstwie nabłonkowej. Praca przedstawia komórki nabłonkowego jelita jako współpartnerów jelitowej zbiorowości immunologicznej regulująych funkcje leukocytów błony śluzowej tkanki jelitowej.

    Streszczenie angielskie: In addition to the barrier function, the intestinal epithelial vells (IECs) play a board role in regional immunologic function. Intestinal epithelium participates in both the innate and acquired immune responses. IECs function as direct participants in innate immunity through the secretion of a variety of factors that contribute to functional barrier formation. IECs are also involved in acquired immune responses through the regulation of local immunologic functions of intraepithelial lymphocytes (IELs) and the subjacent lymphoid tissue. IECs act as the first potential antigen-presenting cell type to come in contact with luminal antigen. In the presence of infection and inflammatory mediators, IECs increase the expression of cytokines and adhesion molecules. While cytokines recruit and activate immune/inflammatory cells, adhesion molecules anchor them to the site of infection. Bidirectional interactions between IECs and IELs play also a role in lymphocyte homing in the intestinal mucosal tissue. This review presents the epithelial cells as members of the intestinal immunologic community regulating the functions of leukocytes within the intestinal mucosa.

    Streszczenie polskie: W pracy przedstawiono korelację pomiędzy ekspresją mRNA histonów H2B i H4 - markerów proliferacji komórek oraz mRNA receptorów kinin B1 i B2 - markerów stanu zapalengo. Wykazany związek pomiędzy ekspresją tych genów wskazuje na udział obu grup genów w procesie nowotworzenia.

    Streszczenie angielskie: Objectives: Histones are involved in process of proliferation and differentiation of the cells. In many lesions the expression of mRNA of histones is a marker of proliferation. The expression of kinin receptors B1 and B2 coexist with inflammatory processes, but in some studies the expression of these receptors was found to influence the growth and differentiation of different cells. Design: In search of proliferation markers in squamous cell vulvar cancer we have analysed the correlation between the expression of the mRNA of histone H2B and H4 and mRNA of kinin receptors B1 and B2. Materials and Methods: The specimen obtained from 46 women operated for squamous cell vulvar cancer stages I to IV according to FIGO were analysed. RNA was extracted and the number of mRNA copies were assessed by QRT-PCR using ABI PRISM TM 7700 (TaqMan). Results: The results obtained in this study indicate a statistically signifciant correlation between mRNA B1 and mRNA H4 (p 0,01), B2 and histone H4 (p 0,01) B1 and histone H2B (p 0,01). Conclusions: The expression of mRNA of histones and kinins receptors may reflects the dynamics of neoplastic growth in vulvar cancer.
