Some clinical and laboratory studies that industrial solvents such as toluene, styrene, xylene, trichloroethylene and carbon disulfide or their mixtures may affect the inner ear, although the mechanism of this process is still not well understood. The aim of this investigation was to assess the incidence of hearing and vestibular disorders (using modern audiological and vestibular tests) in 61 workers exposed to a mixture solvents at the production of paints and varnishes; the control group included 40 age-matched non-exposed subjects. Environmental and biological monitoring revealed that the most significant exposure can be attributed to the following mixture constituents: ethylbenzene, xylene and trimethylbenzene isomers such as pseudocumene, mesitylene and hemimellitene. Electronystagmographic examinations showed the symptoms of vestibular dysfunction, as well as the decreased duration, amplitude and slow phase angular velocity of induced nystagmus in 47.5 p.c. of the subjects exposed versus 5 p.c. of controls. This was accompanied by sensorineural high frequency hearing loss, identified by means of pure tone audiometry in 42 p.c. of those exposed versus 5 p.v. controls, and reduced amplitudes of transiently evoked and distoration-product otoacoustic emissions. The findings closely correspond with the rate total exposure to the solvent mixture. A possible mechanism responsible for ototoxicity of solvents is discussed.

The objective of this study was to evaluate the toxicokinetics and metabolism of pseudocumene after inhalation exposure. Male Wistar rats were exposed to pseudocumene vapors at nominal concentrations of 25, 100 or 250 ppm in the dynamic inhalation chambers for 6 h. Blood samples were collected during (between 1st and 6th h) and after exposure (between 6th min and 6th h). Blood concentrations of pseudocumene were estiamted by gas chromatography using the headspace technique. During a six-hour exposure, the concentration of pseudocumene in blood increased rapidly within the first 2 h reaching then a plateau. The elimination of pseudocumene from blood followed an open two-compartment model. Urine samples were collected from the exposed animals, and metabolites were analyzed by gas chromatography with a flame ionization detector. Three metabolites were measured in the rat uring after hydrolysis: 3,4-dimethylbenzoic acid (3,4-DMBA), 2,4-dimethylbenzoid acid (2,4-DMBA) and 2,5-dimethylbenzoic acid (2,5-DMBA). A significant linear correlation was found between the level of exposure and the concentration of dimethylbenzoic acids. The enzyme kinetics of pseudocumene biotransformation was calculated by Lineweaver-Burk equation. Metabolic constants, Km (mg/l) and Vmax (mg/h/kg), the parameters for pseudocumene biotransformation by rats were estiamted (3,4-DMBA-Km = 28, Vmax = 96; 2,4-DMBA - Km = 7, Vmax = 25; 2,5-DMBA - Km = 7, Vmax = 23).

The distribution, excretion and metabolism of 1,4-dibromobenzene (1,4-DBB) and 1,2-dibromobenzene (1,2-DBB), following a single intraperitoneal administration to female Wistar rats, were investigated using radiotracer 3H and GC-MS technique. The maximum level of 3H after 1,4-DBB administration was detected in all examined rat tissues between 4 and 24 h following the injection. The highest concentrations of 3H were found in fat tissue, muscles, adrenal glands and sciatic nerve. About 50 p.c. of administered dose was still retained in the rat 72 h after injection. For 1,2-DBB, the highest level of 3H was in the liver, kidneys and fat tissue 4 and 8 h after administration. Three days after injection, less than 2 p.c. of the given dose was retained in the rat body. Urine turned out to be the main route of 3H excretion following the injection of both compounds (30 p.c. and 82 p.c., after 1,4-DBB and 1,2-DBB, respectively), and about 4 p.c. of the given dose was excreted in feces. In urine of rats the following substances were indentified (in sequence 1,4-dBB and 1,2-dBB): (1) unchanged parent compounds (5 and 11 p.c.); (2) dibromophenols (84 and 73 p.c.); (3) dibromothiophenols (5 and 10 p.c.) and (4) monobromophenols (1.9 and 0.7 p.c.). This study suggets that 1,2-DBB is characterized by a relatively high turnover rate, whereas 1,4-DBB shows a tendency for long-term retention in the body.
