The biotransformation of endomorphin-1 (EM-1) by brain synaptic membranes has ben studied. Peptide fragments of EM-1 that were formed during incubation with membrane preparations in vitro were isolated by High-performance liquid chromatography (HPLC) and characterized using determination of amono acid composition. At pH 7.4, two degradation products, EM-1(1-3) and EM-1(1-2), were identified. EM-1 was degraded 77.5 p.c. at 30 min incubation with synaptic membranes. The time course of the experiments and the effect of carboxypeptidase inhibitor (CPI) demonstrated that the proteolysis reaction involves the participation of carboxypeptidase activity.

Dendritic cells (DCs) are the most potent antigen-presenting cells (APCs) involved in the induction of adaptive immune responses. The presence of DCs in the central nervous system (CNS) and the active participation of the immune system in a variety of neurodegenerative diseases have been demonstrated. This review will discuss recent findings pertinent to DCs and other antigen-presenting cells in the CNS in health and disease states.

Pro-inflammatory potential of lipoteichoic acid (LTA) from Staphylococcus aureus is controversial. Present study was undertaken to examine ability of highly purified and characterized S. aureus LTA to stimulate production of pro-inflammatory cytokines in human leukocytes at both mRNA and protein level, and to study involvement of Toll-like receptors (TLRs) adn CD14 in this response. Purified LTA was administered to whole human blood ex-vivo (or primary adherent monocytes) and cytokine response assessed in plasma by EIA. Cytokine mRNA was measured by RT-PCR on leukocyate subsets isolated following stimulation. To study involvement of specific receptors for LTA signaling, CHO cells transfected with CD14 and/or TLR2, TLR4 were used, as well as antibodies directed against these receptors. Addition of highly purified LTA to a whole blood or primary adherent monocytes elicited a time and concentration dependent release of TNF-ŕ, IL-1á, IL-6 and IL-8. mRNA encoding TNF-ŕ, IL-1á, IL-6 and IL-8. mRNA encoding TNF-ŕ, IL-1á and IL-6 seemed to be accumulated in monocytes and T cells, but not in granulocytes and B cells. Expression of TLR2, but not TLR4, in chinese hamster ovary cells conferred responsiveness to LTA. However, antibodies directed towards TLR2 (clone TL2.1) or TLR4 (clone THA125) failed to inhibit TNF-ŕ release induced by LTA in the whole blood model and in adherent monocytes. In contrast, blockade of the CD14 receptor with MAb18D11 strongly attenuated LTA induced release of TNF-ŕ in both models.

Background: Renal failure in cirrhotic patients is a severe complication and endotoxemia might be involved. We investigated the effect of endotoxin on renal function of cirrhotic rats and the potential protective role of N-acetylcysteine (NAC). Material/Methods: Hepatic cirrhosis was generated in a rat model by carbon tetrachloride. Both cirrhotic and normal rats were insulted by endotoxin intravenously, while another cirrhotic group was pretreated with NAC. Blood urea nitrogen (BUN) and creatinine were assayed eight hours later. The changes in serum tumor necrosis factor-ŕ (TNF-ŕ) were assayed by ELISA. The histological changes in the kidney were observed after hematoxylinand eosin staining. Results: Endotoxin increased the BUN and creatinine levels in both normal and cirrhotic rats, with a much higher elevation in the latter group. TNF-ŕ concentration was also increase by endotoxin; the changes are positively correlated with BUN and creatinine. NAC pretreatment sigificantly attenuates the effects of endotoxin on BUN, creatnine and TNF-ŕ levels in cirrhotic rats with no improvement in systemic toxicity symptoms. There were no obvious histological changes in the kidney of these animals. Conclusion: Hepatic cirrhosis increased the sensitivity of renal function to endotoxemia, which may be protected by NAC.

This review summarizes recent results investigating the role of certain cytokine gene polymorphisms, including those of TNF-ŕ, IFN-ç, IL-6, IL-10 and IL-1 receptor antagonist (IL-1Ra), in allogeneic stem cell trasplantation. It discusses their role in predicting outcome and the development of a genetic risk index for graft versus host disease (GvHD) in HLA-matched sibling transplants. By the comparative use of an in vitro human skin explant model, initial results suggest than certain cytokine gene polymorphisms may be associated with more severe disease.

The authors present the long-term outcome of reimplantation of ureters into the colon cap (RUCC) during sigmoidocystoplasty (SCP) for the treatment of vesicoureteric reflux (VUR) associated with neurogenic bladder. A retrospective review of thirteen patients with VUR associated with neurogenic bladder (24 ureters) that had RUCC during SCP at our institution was dne. Mean age at surgery was 10.9 yrs (range: 2.1 - 16.4 yrs). Mean follow-up period is 11.5 yrs (range: 5.3 - 16.3 yrs). Severrity of VUR was grade V in 4/24 ureters, grade IV in 12/24, grade III in 4/24, and grade II in 4/24. RUCC was performed into the taenia coli located in the anterior wall of the colon cap in 11/13 patients, and into the posterior wall of the colon cap in 2/13. The average length of the submucosal tunnel created during reimplantation was 23.0 mm. Excisional tapering of the ureter was performed in six ureters, and folding of the ureter in three. Postoperataive complications encountered were vesicoureteric junction stenosis, vesical calculi, and sponttaneous bladder perforation. Renal scarring that progressed to chronic renal faailure developed post-SCP in two patients. There has been no recurrence of VUR. RUCC during SCP is successful and should be actively considered when there is VUR associated with neurogenic bladder and the bladder wall is unsuitable for conventional reimplantation.

Lipoblastoma is a rare pediatric adipose tissue tumor that has to be distinguished from lipoma and myxoid liposarcoma. Cytogenetic studies can help in the differential diagnosis. Most lipoblastoma cases show cytogenetic alterations involving chromosome 8. We review the literature and report a case of a lipoblastoma in a 9-month old male infant with t(2;12)(q32;q15) translocation. According to our knowledge this is the first reported case of lipoblastoma with this translocation.

We report that the intraperitoneal injection of Clostridium perfringens alpha-toxin into mice induces ascites. This phenomenon was monitored by measuring fluid volume and analyzing hematologic data. The mouse toxicity test provides a simple and useful model for examining C. perfringens alpha-toxin-induced vascular permeability.

Background: The present study investigated the contribution of ŕ1-adrenergic blockade to hypertension induced by long-term blockade of nitric oxide and chronic treatment with deoxycorticosterone acetate (DOCA), which produced a low renin model of hypertension. We studied the effects of chronic administration of prazosin, an ŕ1-receptor antagonist, on blood pressure (BP), renal injury, and other variables in Nomega-nitro-L-arginine methyl ester (L-NAME) and L-NAME+DOCA hypertensive rats. Material/methods: The rats were divided into 6 groups: Control, DOCA, L-NAME, L-NAME+DOCA, L-NAME+prazosin, and L-NAME+DOCA+prazosin. Tail systolic BP was measured twice a week. After a 6-week evolution, mean arterial pressure (MAP) was measured, along with selected metabolic, morphological and renal variables. Results: The final MAP values were 105 ń 1 for Control, 107 ń 0.6 for DOCA, 153 ń 3 for L-NAME, 175 ń 2 for L-NAME+DOCA, 126 ń 2 for L-NAME+prazosin and 127 ń 5 for L-NAME+DOCA+prazosin. Proteinuria and hyaline arteriopathy were prevented in L-NAME+prazosin rats and markedly attenuated int eh L-NAME+DOCA+prazosin group. Plasma urea and creatinine were significantly increased in teh L-NAME+DOCA group, but not in the L-NAME+DOCA+prazosin group as against controls. The DOCA and DOCA+L-NAME groups showed relative renal and cardiac hypertrophy, which was not observed in teh DOCA+L-NAME+prazosin group. Cocnlusions: ŕ1-adrenergic tone plays and important role in the increased BP and renal injury of L-NAME hypertension. Our results also indicate that when PRA is suppressed by DOCA in L-NAME hypertension, the increased BP and renal injury are largely dependent on the ŕ1-adrenergi tone.
