    Streszczenie polskie: Wprowadzenie. W podwzgórzu histamina wpływa na różne funkcje, m.in. na spożywanie pokarmu. Betahistyna (antagonista receptora H3 i agonista receptora H1), podana dootrzewnowo (i.p.), w sposób zależny od dawki obniża spożycie pokarmu znacząco statystycznie. Cel pracy. Ocena wpływu betahistyny podanej dożołądkowo, samej oraz jednocześnie z omeprazolem, na spożycie pokarmu. Prawdopodobnie wzrost wydzielania kwasu solnego po podaniu betahistyny hamuje jej działanie ośrodkowe. Omeprazol hamuje wydzielanie soku żołądkowego. Materiał i metody. Doświadczenie przeprowadzono na czterech grupach szczurów, samcach szczepu Wistar. Pierwsza grupa to szczury niegłodzone, druga - przyzwyczajone do "smacznej" diety, trzecia - głodzone jednorazowo przez 20 godnin i czwarta - głodzone po 20 godzin przez 10 dni. We wszystkich grupach betahistyna była podawana w dawce 48 mg/kg dożołądkowo, a omeprazol w dawce 150 ćg/kg podskórnie. Spożycie pokarmu oceniano po 1, 2, 4 i 24 godzinach po podaniu betahistyny w pierwszej grupie szczurów, po 1, 2 i 4 godzinie w grupach szczurów głodzonych oraz po godzinie w grupie szczurów przyzwyczajonych do "smacznej" diety. Wyniki. We wszystkich grupach betahistyna podana dożołądkowo w dawce 48 mg/kg nie miała wpływu na pobór pokarmu. Wnioski. W przeciwieństwie do poprzednich badań, w których betahistyna podana dootrzewnowo znacząco obniżała spożycie pokarmu, w obecnym doświadczeniu po podaniu dożołądkowym nie wykazywała takiego działania, nawet po ...

    Streszczenie angielskie: In the hypothalamus histamine influences various functions, such as feeding. Betahistine (H3-receptors antagoist and H1-receptors agonist), given intraperitoneally (i.p.), significantly decreased food intake in rats in a dose-dependent manner. Objectives. The aim of this study was to assess the influence of betahistine administered intragastrically, alone or with a proton pump inhibitor (omeprazole) on the inhibition of food intake. The stimulation of gastric acid secretion by betahistine inhibits central anorectic activity of this drug. Omeprazole inhibits secretion of hydrochloric acid. material and methods. The experiments were carried out on four groups of male Wistar rats. The first one was on a normal diet, the second was accustomed to "tasty" food, the third was fasted for 20 hours and the fourth group was fasted for 20 hours for ten days. In all examined groups betahistine was administered at a dose of 48 mg/kg intragastrically and omeprazole at a dose 150 ćg/kg subcutanecously. The food consumption was measured after 1, 2, 4 and 24 hours afte betahistine administration in the group of rats on normal diet, after 1, 2 and 4 hours in the group of rats fasted (both methods) and after 1 hour in the group accustomed to "tasty" food.     Streszczenie polskie: Histamina występuje w wielu tkanakch organizmu i wywiera swoje działanie poprzez trzy typy receptorów histaminowych: H1, H2 i H3. W tkankach obwodowych zmagazynowana jest głównie w komórkach tucznych (MC), których obecność wykazano również w macicach ssaków. W macicy ludzkiej większość MC zlokalizowana jest w pobliżu komórek mięśni gładkich, co może wskazywać na ich rolę w odnowie tkanki macicznej podczas cyklu miesiączkowego. Ilość i aktywność MC jest związana ze stanem hormonalnym organizmu.

    Streszczenie angielskie: Histamine isolated from many different tissues, acts via three types of histamine receptors: H1, H2 and H3. In peripheral tissues histamine is mainly stored in mast cells (MC). Presence of mast cells was proved also in mammals' uteri. In human uterus the majority of mast cells ate located close to smooth muscle cells. It might indicate that MC plays a role in tissue remodelling during the menstrual cycle. The quantity and activity of mast cells is in connection with hormonal status of the organism. Although there are some differences, human uterine mast cells are similar to the mast cells isolated from other tissues. It is suggested that histamine is important for normal ovulation, blastocyst implantation, placental blood flow regulation, lactation and contractile activity of uterus. Histamine may also play a role in pathological processes such as pre-eclampsia or preterm delivery. The participation of mast cells and histamine in blastocyst implantation is very controversial. In W/Wv mice (without mast cells) normal implantation was observed. It denies the main role of mast cells in this process but dos not exclude histamine action. In mice the major source of histamine are uterine epithelial cells during early pregnancy. The influence of cytopkines on blastocyst implantation and the role of histamine in cytokines release from the uterine mast cells are also very unclear.

    Streszczenie polskie: Obecność komórek tucznych wykazano nie tylko w macicy, ale również w łożysku. Wydzielane przez te komórki mediatory mogą odgrywać rolę w utrzymaniu prawidłowego przepływu krwi przez łożysko, regulacji ciśnienia tętniczego czy też mieć znaczenie w procesach patologicznych takich jak stan przedrzucawkowy. Sugeryje się również udział histaminy pochodzącej z łożyskowych komórek tucznych w regulacji czynności skurczowej macicy podczas prawidłowego porodu o czasie jak i porodu przedwczesnego. Istnieje bowiem korelacja pomiędzy ilością histaminy w łożysku a występowaniem (bądź nie) akcji skurczowej. Histamina kurczy fragmenty tkanki macicznej większości ssaków poprzez aktywację receptorów H1, ale u niektórych gatunków np. szczur, dominującą odpowiedzią macicy na histaminę jest jej rozkurcz (poprzez aktywację receptorów H2). U ludzi dominującą reakcją macicy na histaminę jest skurcz. Rozkurcz fragmentów ludzkiej tkanki macicznej można uzyskać dopiero po zablokowaniu receptorów H1, chociaż niektórzy agoniści receptora H2 (dimapryt) indukują rozkurcz bez wcześniejszego zablokowania receptorów H. Stan hormonalny (dominacja estrogenów lub progestagenów) również ma wpływ na aktywność skurczową macicy. Nie jest wyjaśniona do końca ani obecność receptorów H3 na włóknach mięśnia macicy ludzkiej, ani ich ewentualna rola w regulacji czynności żeńskiego narządu rodnego.     Streszczenie angielskie: The presence of the mast cells was confirmed not only in the uterus but also in the placental tissue. Mediators released from the placental mast cells may play a role in regulation of placental blood flow and normal blood pressure. Processes such uptake and clearance of vasoactive mediators may be upset in those women who developed pre-eclampsia. Histamine released from the placental mast cells may be involved in the mechanism controling myometrium contractility during the labour at term and preterm delivery. There is a correlation between the level of placental histamine and presence (or not) uterus contractility. Histamine produce a contractile response in isolated myometrial strips, in the majority of mammals, via H1 histamine receptors activation, but in some species e.g. rat, predominant response of uterus is relaxation (via H2 histamine receptors activation). Predominant response of the human uterus to histamine is contaction. Relaxation of human myometrial strips may be evoked after earlier usage of H1 receptors antagonists, although some H2 receptors agonists (e.g. dimaprit) induce the relaxation of human uterus without H1 receptors antagonists. Myometrium contractile activity is under control of sexual hormones. Neither the presence of H3 histamine receptors on the human myometrial smooth cells nor its role in the female reproductive system regulation was proved.
