Streszczenie angielskie: The main goal in the treatment of osteoporosis is to prevent skeletal fractures and to reduce pain. Modern antiresorptive drugs reduce the number of fractures in patients with moderate osteoporosis, they are not able, however, to restore considerably damaged bone structure. Both experimental investigations and prospective clinical trials showed that once-daily injections of parathyroid hormone or its amino-terminal fragment increase the number and activity of osteoblasts, and accelerate the formation of new bone tissue. In postmenopausal women with severe osteoporosis, treatment with amino-terminal fragment of recombinant human parathyroid hormone, given as once-daily subcutaneous injections in a dose of 20 or 40 ćg, resulted in a significant increase in bone mineral density of the spine and femoral neck, without impairing bone qualty. 18-month treatment with PTH decreased by more than 50 p.c. the risk of compression spine fractures and nonvertebral osteoporotic fractures. Parathormone, especially in a dose of 20 ćg daily, proved to be safe and effective in stimulating the reconstruction of damaged bone structure, and decreasing fracture risk inpatients iwth severe osteoporosis.
