    Streszczenie angielskie: Umblical cord blood (UCB) is an alternative source of the haematopoietic stem cells for transplantation. In this study, an effort was made to estimate the medium volume of the collected UCB and conditions of its initial storage before isolation of haematopoietic stem cells. Additionally, the mean contents of CD34+ haematopoietic stem cells among leukocytes, lymphocytes and mononuclear cells was evaluated UCB samples were obtained during deliveries. The collected blood was stored in solutions of studied anticoagulants prepared in studied sulture media, in different time intervals and different temperature. The UCB volume as well as MNC and CD34+ cell counts were correlated with the different perinatological parameters. The concentration, viability and clonogenicity of MNCs were assessed after collection and storage in the studied conditions. The sub-population of CD34+ haematopoietic stem cells within the population of MNCs was isolated using monoclonal antibodies. The collected blood volume depends on newborn weight. Umbilical blood was best stored either in room temperature or in +4řC even for 24 hours in solution made on RPMI with heparin. CD34+ cells concentration correlated only with monomuclear cells counts. The UCB collection and its short-tem storage is a safe and simple method of haematopoietic stem cells recovery for use in transplantation.
