    Streszczenie angielskie: The aim of the study was to assess the risk of asbestos-related malignancies among persons with diagnosed asbestosis. The study covered a cohort composed of 907 men and 490 women afficted by asbestosis, diagnosed in 1970-1997. The follow-up of the cohort continued until 31 December 1999. In all, 421 deaths were registered and causes of death were retrieved for 93.3 p.c. of the deceased. A significantly increased mortality was observed both in the male (300 deaths; SMR = 127; 95 p.c. CI: 113-142) and female (121 deaths, SMR = 150; 95 p.c. CI: 124-179) cohorts. The elevated number of deaths in the male and female cohorts were noted mainly due to respiratory diseases (men: 42 deaths; SMR = 344; 95 p.c. CI: 248-465 women: 20 deaths, SMR = 789; 95 p.c. CI: 482-1219) malignant neoplasms (men: 91 deaths, SMR = 146; 95 p.c. CI: 118-179; women: 34 deaths, SMR = 159; 95 p.c. CI: 110-222), including lung cancer (men: 39 deaths, SMR = 168; 95 p.c. CI: 119-230; women: 13 deaths, SMR = 621; 95 p.c. CI: 331-1062) and pleural mesothelioma (men: 3 deaths, SMR = 2680; 95 p.c. CI: 553-7832; women: 3 deaths, SMR - 7207; 95 p.c. CI: 1031-14612). Taking into account a cumulative dose of fibres, it was found that a significantly increased mortality from lung cancer and pleural mesothelioma applied to persons exposed to a dose above 25 f-y/ml. The results indicate that persons with asbestosis are at higher risk of developing malignant neoplasms, especially lung cancer and mesothelioma.

    Streszczenie polskie: Wstęp. Preparat Prostamer stanowi połączenie w stosunku 1: 1 dwóch surowców leczniczy, zaarejestrowanych jako odrębne preparaty: Prostagalu (olej z pested dyni) i Ekogalu (olej z wątroby rekina) i jest polecany jako środek wspomagający leczenie objawów wczesnego stadium przerostu gruczołu krokowego. Celem pracy było przebadanie wpływu tego preparatu oraz jego składników na rozwój i unaczynienie doświadczalnego nowotworu u myszy. Komórki L-1 sarcoma wszczepiano syngenicznym myuszon śródskórnie (test skórnej angiogenezy, odczyt po 3 dniach, weryfikacja wyników przy użyciu programu komputerowej analizy obrazu "Mouse Observer", wykonanego w ramach grantu KBN nr 7TO8AOr016) lub podskórnie (obserwacja wzrostu guzów i pomiar stężenia hemoglobiny w guzie 14. dnia jako miara jego ukrwienia. Myszy karmiono badanymi preparatami przez cały okres obserwacji, w dawkach dobowych 4 lub 8 mikrolitrów. Wyniki. U myszy karmionych Prostamerem guzy pojawiały się później niż w grupie kontrolnej, a ich objętość była istotnie statystycznie niższa. Prostamer podawany w wyższej dawce zmniejszał również odczyn skórnej angiogenezy, indukowany przez śródskórnie wszczepiane komórki nowotworowe. Nie stwierdzano różnic w ukrwieniu guzów 15. dnia po wszczepieniu komórek nowotworowych w stosunku do grupy kontrolnej. Nie obserwowano istotnych statystycznie różnic objętości guzów w odniesieniu do kontroli dla myszy karmionych pojedynczymi składnikami preparatu Prostamer.

    Streszczenie angielskie: Introduction. Prostamer is a combine (1:1) product of Prostogal (pumpkin seed oil) and Ekogal (shark liver oil), which are registered as separate preparations. It is recommended as adjuvant therapyof BPH early stage. The study was aimed at evaluating the impact of Prostamer on growt and vascuularisation in cancer mouse model. L1 sarcoma cells were implanted to syngenic mice intracutaneouslely (cutaneus angiogenesis test, evaluation after 3 days. outcome verivication with computer software for graphic analysis "Mice Observer"performed as a part of KBN 7TO8AO5016 grant) or subcutaneously (Tumour growth follow-up and haemoglobin concentration on 14th day as a blood supply measure). Mice were administered with evaluated preparations in doses of 4 or 8 ćl. Results. Tumour onset was later in mice administered with Prostamer as compared to control group and additionally the tumour volume was significantly lower. High Prostamer dose resulted also in decrease of cutaneous angiogenesis test induced by intracuteneously implanted tumour cells. There wa no difference in blood supply, compared to the control, assessed on 14th day after tumour cells implantation. No significant difference in tumour volume as compared to control(mice feeded with single ingredients of Prostamer preparation) was observed. Cnclusions. In animal model Prostamer in therapeutic doses or twice the therapeutic doses has shown antineoplastic activity, which in case of higher doses might resulted from suppressing of early angiogenesis.

    Streszczenie angielskie: Tyrosine phosphorylation of numerous proteins is one of the earliest events detectable during Fcç receptor-mediated phagocytosis. We demonstrate that IgG-coated particles associated with the surface of macrophages are enriched with numerous rytosine-phosphorylated proteins. During particle internalization the proteins are still associated with particles but their phosphorylation is reduced. Lyn kinase is phosphorylated both at particle binding and internalization steps. The phosphorylated Syk kinase is the major kinase associated with engulfed particles. Immunofluorescent studies confirm spatial and temporal distribution of Lyn and Syk kinases at different stages of phagocytosis. Our data indicate that ligation of Fcç receptors activites Lyn followed by Syk kinase and in the results multimolecular complex of the kinases and several accompanying tyrosine phosphorylated proteins with Fcç receptors is organized leading to local reorganization of actin-based skeleton and particle uptake.

    Streszczenie angielskie: Recent data indicate that phagocytosis mediated by FcçRs is controlled by the Src and Syk families of protein tyrosine kinases. In this study, we demosntrate a sequential involvement of Lyn and Syk in the phagocytosis of IgG-coated particles. The particles isolated at the stage of their binding to FcçRs (4řC) were accompanied by high amounts of Lyn, in addition to the signaling ç-chain of FcçRs. Simultaneously, the particle binding induced rapid tyrosine phosphorylation of numerous proteins. During synchronized internalization of the particle induced by shifting the cell to 37řC, Syk kinase ans Src homology 2-containing tyrosine phosphatase-1 (SHP-1) were associated with the formed phagosomes. At this tep, most of the proteins were dephosphorylated, although some underwent further tyrosine phosphorylation. Quantitative immunoelectron microscopy studies confirmed that Lyn accumulated under the plasma membrane beneath the bound particles. High amounts of the ç-chain and tyrosine-phosphorylated proteins were also observed under the bound particles. When the particles ere internalized, the ç-chain was still detected in the region of the phagosomes, while amounts of Lyn were markedly reduced. In contrast, the vicinity of the phagosomes was heavily decorated with anti-Syk and anti-SHP-1 Abs. The local level of protein tyrosine phosphorylation was reduced. THe data indicate that the accumulation of Lyn during the binding of IgG-coated particles to FcçRs correlated with strong tyrosine phosphorylation of numerous proteins, suggesting an initiating role for Lyn in protein phosphorylation at the onset of the phagocytosis. Syk kinase and SHP-1 phosphatase are mainly engaged at the stage of particle internalization.

    Streszczenie polskie: Stosując połączone metody chromatografii kolumnowej i preparatywnej chromatografii cienkowarstwoej wyizolowano z Thymi extractum fluidum dwa związki. Ich identyfikację przeprowadzono metodami spektroskopowymi: LSIMS oraz UV. Wyizolowane związki to: 5,4'-dihydroksy-6,7,3'-trimetoksyflawon (kirsilineol) oraz 5,4'-dihydroksy-6,7,8,3'-tetrametoksyflawon (8-metoksykirsilioneol).

    Streszczenie angielskie: Column chromatography with preparative thin layer chromatography has been used to isolate two compounds from Thymi extraxtum fluidum. Identification of those compounds has been carried out by spectroscopic methods: LSIMS and UV. Isolated compounds are: 5,4'-dihydroxy-6,7,3'-trimethoxsyflavon (cirsilineol) and 5,4'-dihydroxy-6,7,8,3-tetramethoxyflavon (8-methoxycirsilineol).

    Streszczenie angielskie: The tradition of therapeutic use of valerian (V. officinalis L.) rhizomes and roots dates back to the times of Dioscorides and Hipocrates, the famous ancient physicians. It is only in the 20th century that the sedative and sleep-improving capabilities of these roots were found to be due to two group sof compounds - valepotriates and sesquiterpenes. The mechanism of their action has not been fully elucidated, but numerous studies conducted on animals allowed to recognize the pharmacological properteies of the individual active compounds of valerian roots and their effect on the central nervous system. Clinical studeis showed the preparations based on extracts from valerian (V. officinalis L.) roots ot be suitable for treating sleep disorders and stress- caused nervous states and revealed their efficacy and safety of application.

    Streszczenie angielskie: EPR method was used to examine on antioxidant activity of black & green teas and also of herbal "teas" so popular in Poland. The content of specific polyphenols - tannins, flavonoids, anthocyanins, procyanidins, phenolic acids and alkaloid - caffeine were determined in examined teas. It was found that following teas possess the strongest antioxidant qualities: Stassen Zielona, Qualitea Natural Green, Zielona z Dalekiego Wschodu, Czarna Cejlońska and Lipton. The largest amount of caffeine was found in: Czarna Cejlońska, Zielona z Dalekiego Wschodu, "Karadeniz" Czarna Turecka, Tetley Earl Grey and Stassen Zielona.

    Streszczenie angielskie: Plasma cell leukemia (PCL) is a very rare variant of multiple myeloma (MM) occuring in about 2 p.c. of newly diagnosed patients. Plasma cell leukemia may develop during the course of MM (secondary PCL) or it can occur without any prior sign of MM (primary PCL). We report a case of aggressive primary PCL with unusual clinical, cytogenetic and molecular features. A 36-year-old male patient was first seen because of fever and bone pain. On the skin of his chest, back, abdomen, and palpebras, there were nodular infiltrations resembling urticaria. White blood cell count was 10.8x10**9/1 with 41 p.c. plasmacytes. Bone marrow aspiration was hypercellular, 93.5 p.c. of cells were atypical plasmacytes and plasmablasts. The cytogenetic analysis of G-banded chromosomes in bone marrow cells yielded the trisomy 8. The skin biopsy specimen showed intensive infiltrates of uninucleated blastic cells similar to those found in the bone marrow. Immunophenotyping of bone marrow and skin neoplastic cells showed CD45+, CD45R0+, CD68+, CD38+ and cytoplasmic kappa light chain+. The neoplasic cells stained negatively for lambda light chain, CD3, CD20, CD30, EMA, CD15, CD34, CD56 and factor VIII. The pattern of IgL genes rearrangement in the bone marrow aspirate, peripheral blood mononuclear cells, and skin specimens was examined by PCR analysis. All studied specimens showed three different Igkappa gene configurations suggesting that the neoplastic cells originated as a result of oligoclonal lymphoproliferation process. The patient received two courses of VAD (vincristine, doxorubicin, dexamethasone)...

    Streszczenie angielskie: Aplastic anemia (AA) may sometimes precede the diagnosis of acute lymphoblastic leukemia (ALL) in children. Such presentation of ALL is externally rare in adults and until now only few such cases have been reported. We present a 40-year-old male with ALL common type, which developed 14 months after the diagnosis of severe AA, successfully treated with corticosteroids. ALL was treated with standard induction chemotherapy but remission has not been achieved. The patient died 6 weeks after the diagnosis of ALL because of central nervous system bleeding. The pattern of IgH gene rearrangement analyzed with PCR method in bone marrow from the period of AA diagnosis and in peripheral blood mononuclear cells from ALL diagnosis showed two different monoclonal IgH configurations as the results of biallelic monoclonal rearrangement of IgH genes. The observed bands in both specimens were identical and indicated that leukemic cells originated from B-cell progenitor were also present in the bone marrow when AA was diagnosed. We suggest that molecular analysis of monoclonality in patients with AA may be important for proper selection of the rare cases of ALL first presenting as marrow aplasia.
