    Streszczenie angielskie: The aim of the study was to assess the effect of storage time on the actual thickness of the histological sectons. The study was performed on 5 brains of adult Wistar rats. The most rapid changes occur at the beginning of the storage process, after about one month the dynamics of changes decreases, but still the thickness of the sections diminishes. It is suggested that quantitative analyses should be performed in similar period of time and the critical care should be taken not only of the chemicals and procedures used, but also of the control of environmental factors.

    Streszczenie angielskie: The aim of the study was to assess the effect of age of the animal upon the real thickness of the frozen sections. The study was performed on 19 rabbit brains. The thickness of the frozen sections regardless of their staining is age-dependent. The relation is proportional during the period from 7 to 180 postnatal day and characterizes both immunohistochemical as well as cresyl violet-stained sections; moreover, changes of the section thickness proceed parallely. It is suggeted that especially for some stereological parameters all required procedures should be standardized to achieve comparable and unbiasedly itnerprtable results.

    Streszczenie angielskie: Immunohistochemical study of the cholinergic innervation of the parvalbumin- ad calbindin-containing cells in the hippocampus was conducted on 30 rat brains of various postnatal ages: PO, P4, P7, P14, P21, P30, P60 and P180. Sections with double immunostaining for vesicular acetylcholine transporter (VAChT; the marker of cholinergic cells, fibres and terminals) and parvalbumin (PV) or calbindin (CB) were analysed using confocal laser-scanning microscope. Obtained data demonstrate that the pattern of cholinergic innervation of calbindin- and parvalbumin-immunoreactive hippocampal neurones shows some differences. During development as well as in the adult species cholinergic terminals preferentially innervate CB-containing neurones, while cholinergic terminals on PV-containing cells were observed rarely. Cholinergic endings on the CB-ir neurones are localised both on their somata and dendrities, whereas on PV-ir cells they form synaptic contact predominantly with processes. In spite of the unquestionable cholinergic influence particularly on CB-ir cells, the number of cholinergic endings suggests that this input seems not to be crucial for the activity of the studied cell populations.

    Streszczenie angielskie: We have demonstrated for the first time that the steroid metabolite, 2-methoxyestradiol (2-ME) is a powerful growth inhibitor of human osteosarcoma 143 B cell line by pleiotropic mechanisms involving cell cycle arrest at two different points and apoptosis. The ability of 2-ME to inhibit cell cycle at the respective points has been found concentration dependent. 1 ćM 2-ME inhibited cell cycle at G1 phase while 10 ćM 2-ME caused G2/M cell cycle arrest. As a natural estrogen metabolite 2-ME is expected to perturb the stability of microtubules (MT) in vivo analogously to Taxol - the MT binding anticancer agent. Contrary to 2-ME, Taxol induced accumulation of osteosarcoma cells in G2/M phase of cell cycle only. The presented data strongly suggest two different mechanisms of cytotoxic action of 2-ME at the level of a single cell.

Sections with double immunostaining for vesicular acetylcholine transporter (VAChT; the marker of cholinergic cells, fibres and terminals) and calretinin were analysed using confocal laser-scanning microscope. Obtained data demonstrate that during development as well as in adult species calretinin-containing neurones in teh rat hippocampus form sparse synaptic contact with VAChT-ir terminals. It seems probable that cholinergic innervation is not crucial for the functioning of CR-ir cells - probably they remain under the greater influence of a system other than the cholinergic system.

    Streszczenie angielskie: In the present study we examined the effects of menadione, a redox cycling agent, on structural changes of human osteosarcoma line 143B cells. It has been previosly reported that menadione can cause necrotic or apoptotic cell death in a concentration- depending manner. In our experimental model, cells were treated with 100 ćM menadione for 24 hours. Using electron microscopy technique cells carrying three kinds of morphological changes were detected: necrotic cells, apoptotic cells and those demonstrating a co-existence of apoptotic and necrotic features in one single cell.

    Streszczenie angielskie: In the present study we have shown that quinolinic aromatic aminoxyls are very efficient in protecting lipids of endoplasmic reticulum membranes against hydroperoxide-induced oxidation. The efficacy of these aminoxyls as protectors of lipids was much higher than the water-soluble 4-OH-TEMPO. We have also shown that QAL causes distinct changes of the morphology of mitochondria: from filamentous to granular enlarged structure via the folding of the former. QAL induces also perinuclear clustering of mitochondria. C-QAL as well as 4-OH-TEMPO treated cells revealed filamentous and scattered pattern of mitochondria. Antioxidant activity of QAL as well as morphological changes of mitochondrial raise the possibility that this drug can affect cell physiology via changes of mitochondrial function.
