Against many viral diseases caused for example by HSV, EBV, CMV, HIV, RSV, HCV for which vaccines are not available, chemiotherapeutics seem to have the principal significance. High progress in development of new antiviral compounds is observed. In addition to synthetic compounds a large number of naturally occuring substances have been shown to posses antiviral activity. One of such substances is tannic acid. In this study comparison of antiviral activity of tannic acid, acyclovir (ACV) and ganciclovir (GCV) against cytomegalovirus (CMV) is presented. The MRC5 cells infected with CMV and treated with different compounds were analyzed by flow cytometry and cythopatic effect inhibition test for inhibition of virus replication and by MTT assay for cytotoxity. It has been shown that tannic acid has antiviral activity against cytomegalovirus and that expression of virus antigens measured as median fluoroscence intensity (MFI) by flow cytometry can be used for evaluation of virus replication.

It is currently believed that oxidative stress and inflammation play a significant role in atherogenesis. Artichoke extract exhibits hypolipemic properties and contains numerous active substances with antioxidant properties in vitro. We have studied the influence of aqueous and ethanolic extracts from artichoke on intracellular oxidative stress stimulated by inflammatory mediators (TNFŕ and LPS) and ox-LDL in endothelial cells and monocytes. Oxidative stress which reflects the intracellular production of reactive oxygen species (ROS) was followed by measuring the oxidation of 2', 7'-dichlorofluorescin (DCFH) to 2', 7'-dichlorofluorescin (DCF). Agueous and ethanol extracts from artichoke were found to inhibit basal and stimulated ROS production in endothelial cells and monocytes in dose dependent manner. In endothelial cells, the ethanolic extraxt (50 ćg/ml) reduced ox-LDL-induced intracellular ROS production by 60 p.c. (p 0,001) while aqueous extract (50 ćg/ml) by 43 p.c. (p 0,01). The ethanolic extract (50 ćg/ml) reduced ox-LDL-induced intracellular ROS production in monocytes by 76 p.c. (p 0,01). Effective concentrations (25-100 ćg/ml) were well below the cytotoxic levels of the extracts which started at 1 mg/ml as assessed by LDH leakage and trypan blue exclusion. Penetration of some active substances into the cells was necessary for inhibition to take place as juged from the effect of preincubation time. These results demonstrate that artichoke extracts have marked protective properties against oxidative stress induced by inflammatory mediators ans ox-LDL in cultured endothelial cells and monocytes.

The purpose of this work was to evaluate the relationship between pathological changes and presence of CMV DNA in atherosclerosis plaques in wall of coronary vessels. The presence of CMV DNA was investigated by nested PCR in atherosclerotic lesions and unchanged site of a vessel taken postmortem from the same person. Also the serum samples were investigated for the presence of anti-CMV-IgG and IgM antibodies by ELISA and CMV DNA by nested PCR. Intensity of atherosclerosis was evaluated by postmortem examination with 5 point scale. Atherosclerotic changes with different intensity were observed in 30 out of 35 examined persons. CMV DNA was found in 30 p.c. of vessels with lesion, but was not in arterial walls of patients without atherosclerosis (grade O). Intensity of atherosclerotic changes was correlated with age and anti-CMV IgG level, but small number of cases with high advanced changes (II-IV grade) did not allowed a more precise analysis.

This paper contains review of materials, patients' group and methods used in diagnosis of HCMV in routine practice in Department of Virology, National Instutute of Hygiene. In the period January 2000 - April 2002 altogether 773 cases underwent diagnostic analysis. Analyses of immunocomponent persons tested in order to confirm infection or determine the immunological status, patients after transplantation, and women in the procreative period with suspected HCMV infection constituted 94.1 p.c. of the entire group. The materials from patients were investigated using ELISA, shell vial, OIF, pp65 and PCR methods. The results and diagnostic value of different methods were discussed.
