It has been suggested that metabotropic glutamate (mGlu) receptor agonists selective for Group II mGlu receptors may have antipsychotic action. Therefore, we studied whether the effects, which could be related to psychotomimetic action of hallucinogenic drugs, are inhibited by Group II mGlu receptor agonists. The selective mGlu2/3 agonist LY354740 and LY379268 inhibited (ń)1-(2,5-dimethoxy-4-iodophenyl)-2-aminoproprane (DOI)-induced head twitches in mice in a dose-dependent manner. Furthermore, LY379268 suppressed an increase in the frequency of sponataneous excitatory synaptic potentials induced by bath-applied DOI in layer V pyramidal cells recorded in the murine medial frontal cortex. The data indicate that Group II mGlu receptor agonist may counteract the effects of hallucinogenic drugs.

Zn is the second most prevalent trace element in the body and is present in particularly large concentrations in the mammalian brain. Although Zn is a cofactor for many enzymes in all tissues, a unique feature of brain Zn is its vesicular localization in presynaptic terminals, where its release is dependent on neural activity. Synaptic Zn, influencing excitatory glutamate, probably plays a modulatory role in synaptic transmission. This paper reviews glutamatergic system, including ionotropic and metabotropic glutamate receptors, and its role of zinc in the modulation of excitatory transmission.

Previous studied showed that chronic electroconvulsive shock (ECS) or impiramine treatment induced a subsensitivity of group I metabotropic glutamate receptors (mGluR) in hippocampus. In the present study effects of antidepressant treatment on the expression og mGluR1a and mGluR5a, belonging to the group I mGluR, were investigated in rat brain hippocampus using immunohistochemical and Western blot methods, respectively. Male Wistar were treated singly or chronically for 21 days with imipramine, 10 mg/kg, twice daily; with ECS (90 mA, 50 Hz, 0.5 s) every second day; or with haloperidol, 1.2 mg/kg, once daily. Appropriate controls were injected with saline. Rats were sacrificed 24 h after last treatment and their hippocampi were taken out for analysis. It was found that the mGluR1a-immunoreactivity expression increased significantly in Ammon's horn (CA) regions after chronic ECS. The most pronounced effect was observed in the CA3. No significant effects were found after single treatment or after haloperidol. The expression of mGluR5a increased significantly after chronic impiramine in the CA1 and after chronic ECS in the CA3 region. The results obtained indicate an influence of antidepressant treatment on group I mGluR. This increase in the receptor protein level may be a compensatory mechanism developing after chronic treatment.

Several lines of evidence suggest a crucial involvement of glutamate in the mechanism of action of anxiolytic and antidepressant drugs. The involvement of group I mGLu receptors in anxiety and depression has also been proposed. In view of the recent discovery of anxiolytic- or antidepressant-like effects of acute injections of 2-methyl-6-(phenylethyl)-pyridine (MPEP), a selective and brain penetrable mGlu5 receptor antagonist, we designed the present study to examine anxiolytic- and/or antidepressant-like effects of multiple administration of this drug. The anxiolytic-like effects of MPEP were evaluated in rats using the conflict drinking test. The antidepressant-like effect was estimated using the rat olfactory bulbectomy model of depression. Seven subsequent injections of MPEP (1 mg/kg) significantly (by 320 p.c.) increased the number of shock accepted during the experimental session in the Vogel test. MPEP given once daily at a dose of 10 mg/kg, restored the learning deficit of bulbectomized rats after 14 days of treatment, remaining without any effect in the sham-operated animals. N-methyl-D-aspartic acid (NMDA)-induced convulsions in mice were not affected by a single injection of MPEP (30 mg/kg) indicating that at this dose MPEP did not block NMDA receptors. The results indicate that the prolonged blockade of mGlu5 receptors exert anxiolytic- and antidepressant-like effects in rats. No tolerance to anxiolytic-like action occurs. The previously mentioned results further indicate that antagonists of group I mGlu receptors may play a role in the therapy of both anxiety and depression.

The aim of the present study was to find out whether the metabotropic receptor 1 (mGluR1) and group II mGluRs, localized in the striatum, are involved in antiparkinonian-like effects in rats. Haloperidol (1 mg/kg ip) induced parkinsonian-like muscle rigidity measured as an increased resistance of a rat's hind foot to passive flexion and extension at the ankle joint. (RS)-1-aminoindian-1,5-dicarboxylic acid (AIDA; 0.5-15 ćg/0.5 ćl), a potent and selective mGluR1 antagonist, or (2R,4R)-4-aminopyrrolidine-2,4-dicarboxylate (2R,4R-APDC; 7.5-15 ćg/0.5 ćl), a selective group II agonist, was injected bilaterally into the striatum of haloperidol-treated animals. AIDS in doses of 7.5-15 ć/g/0.5 ćl diminished the haloperidol-induced muscle rigidity. In contrast, 2R,4R-APDC injections were ineffective. The present results may suggest that the blockade of striatal mGluR1, but not the stimulation of group II mGluRs, may ameliorate parkinsonian muscle rigidity.

Most conventional antidepressant drugs influence serotoninergic adrenergic, and/or dopaminergic systems, increasing serotonin, norepinephrine and dopamine synaptic availability. More recently has focused on glutaminergic system. Both preclinical and clinical studies, showing antidepressant-like actions of compounds which reduce transmission at N-methyl-D-aspartate (NMDA) receptors, indicate possible involvement of glutamatergic system in the etiology of depression. Since glutamatergic transmission is controlled not only by inotropic but also by metabotropic glutamate receptors (mGluR), their involvement in the etiology and the therapy of depression was also postulated. Recent studies, showing that antidepressant treatment may influence mGlu receptors, together with the findings that group I mGluR antagonists, may possess antidepressant-like action, support this hypothesis.

Recent preclinical data have suggested that glutamate NMDA receptor may be involved in the mechanism of action of antidepressant treatments. Functional antagonists of the NMDA receptor complex exhibit an antidepressan-like effect in animal tests that predict antidepressant activity and in animal models of depression. Zinc, a very potent inhibitor of the NMDA receptor, is active in the forced swimming test in rats and mice. The present study investigated the interaction of zinc with antidepressants in the forced swimming test in mice. Mice were injected with imipramine or citalopram alone and in combination with zinc. Low, ineffective per se doses of imipramine and citalopram administered together with low, ineffective doses of zinc were active in this test. The present data support the notion that inhibition of the NMDA receptor participates in an antidepressant action, and further demostrate particular role of zinc in this activity.

We examined the anxiolytic-like activity of (1S,3R,4S)-1-aminocyclopentane-1,3,4-tricarboxylic acid (ACPT-I) using the Vogel conflict drinking test, while antidepressant-like effects of this compound were evaluated using Porsolt's test. ACPT-I, a selective group III mGlu receptor agonist, produced a dose-dependent anticonflict effect after intrahippocampal injections and antidepressant-like effect in rats after intraventricular injections. These data suggest that selective group III mGlu receptor agonists may become a new class of anxiolytics and/or antidepressants.

Wstęp.Nadmierne spożywanie chlorku sodu uważane jest za główny czynnik ryzyka rozwoju choroby nadciśnieniowej, dlatego też spożywanie mniejszych ilości tego związku zalecane jest zarówno w leczeniu, jak i w prewencji choroby nadciśnieniowej. W dostępnej literaturze znaleźć można jednakże niewiele informacji na temat wpływu obniżonej lub zwiększonej zawartości soli kuchennej w diecie na możliwości wysiłkowe człowieka. Cel pracy. Ocena wpływu diety z różną zawartością chlorku sodu (NaCl) na wydolność aerobową człowieka. Metodyka. U 11 zdrowych mężczyzn, podczas testu wysiłkowego ze stopniowo narastającym obciążeniem, trzykrotnie obserwowano wpływ różnych ilości NaCl na poziom VO2. Analizując wskaźniki oddechowe, uzyskane przy poszczególnych obciążeniach, wyznaczono TDMA i VO2max. Przed I próbą dieta badanych była niezmieniona - średnia wartość kaloryczna przyjmowanych pokarmów wynosiła ok. 2800 kcal/dobę, a zawartość NaCl w diecie ok. 6,8 g/dobę. Próbę II poprzedziło spożywanie przez nich dodatkowo 8 g NaCl dziennie przez 3 dni, a próbę III - stosowanie przez 3 dni diety niskosolnej (1,3 g/dobę). Wyniki. Najwyższe wartości VO2 max (55,88 ń 7,09 ml/kg/min), czasu pracy (17,45 min) i czasu osiągnięcia progu TDMA (12,18 min) zaobserwowano po diecie wysokosolnej, zaś najniższe wartości VO2max (48,94 ń 4,36 ml/kg/min), czasu pracy (15,75 min) i czasu osiągnięcia TDMA (9,88 min) odnotowano po diecie niskosolnej. Introduction. High sodium intake is thought to increase the risk of high blood pressure. Therefore, a reduction in sodium chloride intake is advised, in both the treatment and prevention of hypertension. However, there is insufficient literature available on the subject of the influence of low or high salt intake on physical performance in humans. Purpose. The aim of our study was to assess the influence of diets with a varied NaCl content on human aerobic physical performance. Methods. Changes in VO2 were examined in 11 men who performed an exercise test to exhaustion. Analysis of ventilatory indices at certain levels of workload lead to the determination of TDMA and VO2max. During phase I the test was carried out with the same diet content. The mean calorific value of the daily diet was 2800 kcal and the aomunt of NaCl in the diet was about 6.8 g/d. Prior to engaging in phase II of the exercise test, participants were given diets containning 8 g NaCl per day for 3 consecutive days. Finally, prior to engaging in phase III of the test, a low sodium diet was administered (1.3 g/24 hr). Results. The highest VO2max values (55.88 ń 7.09 ml/kg/min), test duration (17.45 min) and time of reaching TDMA threshold (12.18 min) were observed at phase II.
