FasL molecule expressed on activated T cells induces apoptosis in Fas-expressing cells. It is possible that apoptosis induced by FasL is involved in the process of allograft destruction brought about by infiltrating T cells. The aim of our study was to evaluate expression of FasL gene in peripheral blood T cells of renal allograft recipients (RAR). We have studied 25 patients: 16 with uneventful stable course (RAR-S) and nine during biopsy proven chronic rejection (RAR-CH). The relative expression of FasL mRNA compared with that of á-actin was established by semi-quantitive RT-PCR. We have found that FasL gene expression was significantly increased in T cells of RAR-CH compared to RAR-S (P 0.01). Our results suggest that T cells expression of FasL gene is increased during chronic rejection. Therefore, this phenomenon may pay a role in allograft injury associated with that process. Further studies are needed to unravel possible clinical consequences of observed differences in T cell expression of FasL.

Recent data indicate that human T lymphocytes can adhere to elastin and respond to co-stimulatory signals of that protein. This reactivity is mediated by non-integrin receptor, elastin binding protein. In addition, another receptor belonging to integrin family may be also involved. T cell interactions with elastin (but not other extracellular matrix proteins) appear to be upregulated in healthy males and at least some patients with vasculitis. Interestingly, statins in pharmacological concentrations strongly and selectively block those interactions. Our data point to the potential role of T cell interactions with elastin in immunopathology of vasculitis and atherosclerosis.

Background. Vein graft failure occurs in 15 p.c. of patients during one year after coronary by-pass grafting. The mechanisms of graft occlusion and preventive methods have not yet been well established. Aim. To test the hypothesis that the use of originally developed extremal dacron mesh affects the proliferative and degenarative changes in vein grafts implanted to the cliterial circulation. Methods. In 21 sheep standard cephalic vein graft or hybrid graft consisting of cephalic vein covered with extermal darcon mesh, were implainted to the cartoid artery. The histological and immunocystochemical characteristics of the histologic specimens harvested at various time points from implaimentation were compared. Results. In both types of grafts a gradual intimal and medial thickening was observed. Five days after implantation the mean intimal thickness in the hybrid grafts was 19.2 ń 10.7 um vs 24.4 ń 6.5 um in the non-stented graft, after 9 days - 22.3 ń 5.8 um vs 26.2 ń 10 um, after 14 days - 22.6 ń 7.6 um vs 52.2 ń 15 um, after 6 weeks - 36.6 ń 20.7 um vs 89.9 ń 31 um, after 8 weeks - 56.6 ń 30.8 um vs 103.6 ń 28 um, after 10 weeks - 58.3 ń 20.6 um vs 132.5 ń 31.6, and 12 weeks - 72 ń 21.5 um vs 244 ń 99.8 um, respectively (p 0.001 for all comparisons). The histological assessment in the light microscopy revealed progressive degeneration of the vein wall markedly more pronouced in grafts without rather than extermal stenting. Conclusions. The use of external stenting in a form of dacron mesh covering vein graft implainted in the arterial circulation reduced graft wall hypertrophy, slows down the speed of intimal thickening and inhibits cellular proliferation in the graft wall.

Przygotowano lipopolisacharydy jednego nieenterotoksynotwórczego (NTBF) i trzech enterotoksynotwórczych (ETBF) szczepów B. fragilis oraz enterotoksynę B. fragilis. Badano wpływ polimyksyny B na stymulację ekspresji cząsteczek adhezyjnych: selektyny E, ICAM-1 i VCAM-1 na powierzchni komórek ludzkiego śródbłonka naczyniowego przez przygotowane preparaty bakteryjne. Do oznaczenia cząsteczek adhezyjnych zastosowano test ELISA z przeciwciałami monoklonalnymi. Stwierdzono, że polimyksyna B zmienia poziom ekspresji selektyny E, ICAM-1 i VCAM-1 podczas jednoczesnego działania z lipopolisacharydami lub enterotoksyną B. fragilis.
