    Streszczenie angielskie: Stroke-induced inflammatory reaction leads to the accumulation of leukocytes in the brain ischaemic region, where they exert a detrimental effect - promotion and extension of cerebral damage. Intracerebral infiltration of peripheral blood leukocytes requires prior endothelial-leukocyte interactions that are mediated by such cell surface proteins as adhesion molecules. Among adhesion molecules, it is the immunoglobulin gene superfamily (IgSF) that is responsible for strong attachment and transendothelial migration of leukocytes. The principal members of IgSF are: intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1) and platelet endothelial cell adhesion molecule-1 (PECAM-1). In this review the following issues were described and discussed: an increased expression of ICAM-1 and VCAM_1 in ischaemic brain as well as a detection of their soluble(s) forms in sera of stroke victims. The presented data suggest the involvement of both ICAM-1 and VCAM-1 in the sequence and timing of the infiltration of leukocytes into the brain ischaemic zone after stroke. They have also revealed changes in serum concentrations of sICAM-1 and sVCAM-1 that are characteristic for stroke. Recently, increase in sPECAM-1 levels in serum and cerebbrospinal fluid (CSF) has been shown within 24 h of the onset of stroke, having indirectly suggested involvement of the molecule in the inflammatory events during the early phase of stroke.

    Streszczenie angielskie: Interleukin-15 is a novel cytokine produced by monocytes/macrophages and sharing several biological activities with IL-2. IL-15 induces T cell proliferation, enhances natural killer (NK) cell cytotoxicity and also stimulates B cells to proliferate and secrete immunoglobulins. The purpose of our study was to measure IL-15 levels in the serum and CSF of 21 patients with relapsing-remitting form of MS, 9 with active gadolinium enhancing lesions in MRI, 12 without enhancing MRI lesions and to compare the results with the control group. IL-15 levels were measured by ELISA. We found a significant incrase of IL-15 in the sera of patients with MS in comparison with the control group consisting of 8 patients with tension headache. IL-15 serum levels were highest in patients with active, gadolinium enhancing lesions. IL-15 CSF levels were low and there was no difference between studied groups. The results may suggest the contribution of IL-15 in the immunopathogenesis of multiple sclerosis.

    Streszczenie angielskie: PECAM-1 is an adhesion molecule - a member of the immunoglobulin superfamily -involved in the transendothelial migration of leukocytes PECAM-1 is expressed on lymphocytes, monocytes and granulocytes. It is also found on endothelial cells and platelets. We present data showing that the cell-bound form of PECAM-1 expression on monocytes is increased in MS patients, compared to controls. We also show that PECAM-1 is significantly over-expressed on lymphocytes in patients with active MRI lesions, when compared to those without gadolinium - enhancing lesions. Our results suggest that the cell-bound form of PECAM-1 can be regarded as a marker of MS activity.

    Streszczenie angielskie: Inflammatory response following acute cerebral ischaemia exacerbates damage and enlarges infarct extent. The role of adhesion molecules mediating attachment and migration cells within the ischemic cerebrovascular region seems to be critical in the development of stroke-induced inflammatory reaction. Platelet endothelial cell adhesion molecule-1 (PECAM-1) present at cell to cell junction is reguired for transendothelial migration of leukocyters. We have recently demonstrated that ischaemic stroke patients displayed increased soluble PECAM-1 isoform (sPECAM-1) levels in serum and cerebrospinal fluid (CSF) within 24 after the disease onset. The aims of the present study performed in the same group of 23 ischaemic stroke patients were to investigate the relationship between the levels of sPECAM-1 and the extent of early brain damage as well as stroke severity and its outcome. We have compared sPECAM-1 levels in serum and CSF samples obtained from the patients within 24 h of first-ever completet hemispheric ischaemic stroke with the volume of anatomically relevant rarly computed tomography (CT) of the brain hypodense areas as well as the neurological stroke severity estimated according to the Scandinavian Stroke Scale (SSS) scores and the functional disability assessed with the Barthel Index (BI) scores.     Streszczenie angielskie: Proinflammatory cytokines produced by Th-1 cells and cytokines with immunosupressive properties play an important role in the pathogenesis of multiple sclerosis (MS). Glatiramer acetate (GA) is one of the most important immunomodulatory agents used in the therapy of MS. The mechanism of action of GA in MS is not yet fully explained. In our previous study we found the significant down-regulation of interleukin 18 (IL-18), a proinflammatory cytokine inducing the production of interferon ç during therapy wiht GA in MS patients. The purpose of this study was to evaluate the effect of GA in a dose 20 mg daily in a period of 6 months on interleukins IL-10 and IL-12. Thirty-one patients with definite MS and 30 control subjects were the subject or our study. The interleukins levels in sera were measured by the ELISA test. A significant increase was found of IL-12 and also of IL-10 levels in MS patients in comparison with control group. We also established a significant decrease of IL-12 after 3 and 6 months of GA therapy and some insignificant differences in the level of IL-10 (the decrease after 3 months and the increase after 6 months). IL-12 is proinflammatory cytokine secreted by blood mononuclear cells, including dendritic cells in response to antigens and mitogens and is thought to contribute to the pathogenesiss of MS. Therefore the established downregulation of IL-12 as well as that previously described of IL-18 suggest a marked relationship between the clinical effect and downregulatory action of GA on proinflammatory interleukins.
