Angiogeneza, czyli proces powstawania naczyń może występować zarówno w warunkach fizjologicznych, jak i w patologicznych, np.: gojeniu ran, nowotworach, chorobie niedokrwiennej serca lub naczyń, przewlekłych zapaleniach. Angiogeneza nowotworowa odgrywa bardzo istotną rolę zarówno w ich rozwoju, jak i w powstawaniu przerzutów. Proces ten jest zainicjowany przez niedotlenienie komórek nowotworowych, mutacje genów supresorowych i onkogenów. Komórki nowotworowe aktywują swoiste czynniki angiogenne przede wszystkim naczyniowo-śródbłonkowy czynnik wzrostu (VEGF), które stymulują migrację i podziały komórek śródbłonka. Poznanie czynników proangiogennych i antyangiogennych pozwoli prawdopodobnie ingerować w proces angiogenezy w celu jej zahamowania (w nowotworach) albo pobudzenia w leczeniu np. choroby niedokrwiennej serca. W artykule przedstwaiono niektóre aspekty aktualnej wiedzy na temat angiogenezy i jej regulacji.

The pathogenesis of thyroid cancer is complex. Iodium deficiency, genetic factors, sex, older age, irradiation in childhood, thyroid growth stimulating antibodies and epithelial growth factor possibly affect its development. Recent reports, also epidemiological, have shown that differentiated thyroid cancer (papillary, follicular, oxyphillic) is dependent on sex hormones, especially estrogens. This has prompted research into the presence of estrogens and progesterone, as well as androgen receptors (ER, PR and AR) in normal and neoplastic thyreocytes and estradiol content in thyroid tissue. The results of these investigations imply that thyroid cancer are estrogen-dependent. There is, however, no agreement in reports about correlation between tumor malignancy and ER, PR and AR expression. In this study 72 thyroid glands have been examined using immunohistochemical assays with ERs and PRs antibodies. Positive staining was demonstrated mainly in differentiated thyroid cancer nuclei (17 ERs positive and 2 PRs positive). There was no expression of antibodies in non-neoplastic cells or in adjacent tissues. The nuclei of 3 oxyphillic and 4 follicular adenomas were also ERs positive. On the other hand nuclei of 5 fokkicular adenomas were PRs positive. These results are in agreement with most available data. It seems, however, that the material so far examined by the investigators is too small. Furthermore, because of using various methods the results cannot be compared. Further studies are necessary to reveal if there is any true influence of sex hormones on the development of thyroid lesions and if the detection of sex hormone receptors may help in choosing adequate therapy.

Vascular endothelial growth factor (VEGF) is one of the factors related to angiogenesis of tumors and physiological and pathological proliferative processes. Location of VEGF in human thyroid tissue with various thyroid disorders was studied in order to explore its possible involvement in proliferative processes. Immunohistochemical examination was performed on 69 formalin-fixed, paraffin-embedded thyroid tissue specimens using the labelled streptoavidin biotin peroxidase complex detection system. VEGF was not identified in normal thyroid follicular cells. Some but not all tumor thyreocytes expressed VEGF in cytoplasm. VEGF positive expression was found in 6/15 patients with papillary carcinoma, 4/9 with follicular carcinoma, 14/28 with follicular adenoma and 2/17 with adenomatous goiter. In benign follicular adenoma and adenomatous goiter the weak expression of VEGF was found in small areas of the tumor tissues, whereas in malignant tumors it was found strongly in many cells. VEGF probably functions as a hypoxia-inducible angiogenic factor, and the expression of VEGF is stronger in malignant tumors, which need more oxygen supply to proliferate. An intensive VEGF production by differentiated thyroid carcinomas could be a promising marker of tumor aggressiveness and may also be useful as a predictor of metastatic potential and extension of tumor mass.
