Jedną z jej najciekawszych form jest konstruowanie teorii naukowych, co wskazuje na niebagatelną rolę jaką odgrywa w tym procesie. Twórczość, niemal immanentnie związana z wyobraźnią, staje się granicą tego, jak dalece potrafi posunąć się naukowiec w swych spekulacjach i eksploracjach, aby konkretna teoria wnosiła coś istotnego do naszej wiedzy. Odwaga wyobraźni staje się probierzem innowacyjnych heurystyk i niespetryfikowanego oglądu świata.

    Streszczenie angielskie: Imagination manifests itself in very different forms. One of the most interesting problems there is construction of scientific theory, that shows not trifling function of imagination in this process. Creation, almost limit for research worker in his speculations and explorations, so that his specific theory brings something essential to our knowledge. Courage of imagination becomes gauge of innovative heuristics and not petrified view of world.

    Streszczenie angielskie: Antipsychotic drugs can modulate transcription factors ansd also nuclear receptors, but their action on glucocorticoid receptors (GR)-members of the steroid/thyroid hormone receptor family has been studied so far. In the present study we investigated effects of various antipsychotics on the glucocorticol-mediated gene transcription in fibroplast cells, stably tranfected with a mouse mammary tumor virus promoter (LMCAT cells). Chlorpromazine (3-100 ćM) inhibited the corticosterone-induced gene transcription in a concentration- and time-dependent manner. Clozapine showed a similar, but less potent effect, while haloperiod acted only in high concentrations, and other antipsychotics drugs (sulpiride, raclopride, remoxipride) were without any effect. It was also found that a phorbol ester (an activator of protein kinase C (PKC) and A-23187 (Ca2+-ionophore) attenuated the inhibitory effect of chloropromazine on the GR-induced gene transcription. An antagonist of the L-type Ca2+ channel, as an inhibitor of phospholipase C (PLC) inhibited the corticosterone-induced gene transcription, but had no effect on the chlorpromazine-induced changes. The involvement of a PKC/LC pathway in the chlorpromazine action was confirmed by Western blot analysis showed that the drug in question decreased the PLC-á1 protein level, and to a lesser extent that of the PKC-ŕ protein in LMCAT cells.     Streszczenie angielskie: In order to determine whether the status epilepticus leads to alterations in th e neurosteroid effect on excitatory amino acid transmission, we studied the influence of allopregnanolone on aspartate release and glutamate uptake in mouse hippocampus at various times after kainate administration. No significant differenses in the K+stimulated D-[3H]-aspartate release from the hippocampi of saline- and kainate-treated mice were observed; however, that parameter tended to fall in tissues collected 1h after kainate administration. Allopregnanolone significantly attenuated the K+-stimulated D-[3H]-aspartate release from the hippocampi of control animals, as well at 24 h and 7 days after kainate injection; in contrast it did not affect amino aaacid release from the hippocamp collected 1h after kainate administration. Kainate administration had noeffect on [3H]-glutamate uptake after 1 and 24 h, but elevated that parameter on day 7. Allopregnanolone (10 and 100 ćM) did not affect [3H]-glutamate uptake in control and kainate-treated mice. In conclusion, the present study indicates a loss of the inhibitory effect of allopregnanolone on the potasium - stimulated D-[3H]-asparate release from mouse hippocampus during the kainateinduced status epilepticus; moreover, it excludes involvement of this neurosteroid in the regulation of hipocampal [3H]-glutamate uptake in both control and kainate-treated mice.

    Streszczenie polskie: Wprowadzenie. Padaczka należy do często występujących chorób neruologicznych. W jej przebiegu u 25-30 proc. pacjentów farmakoterapia jest nieskuteczna. Uzasadnione jest zatem poszukiwanie nowych rodzajów leków przeciwpadaczkowych w celu leczenia opornych na leki postaci padaczki i lepszego oceniania skuteczności dotychczas stosowanych terapii. Celem pracy jest krótkie przedstawienie nowych koncepcji i kierunków badawczych w zakresie farmakoterapii padaczki, ze szczególnym uwzględnienim możliwości wykorzystania zdobyczy neurochemii i biologii molekularnej. Omówienie. Ewolucja poglądów na mechanizmy powstawania drgawek i działania leków przeciwpadaczkowych w dużym stopniu odzwierciedla postępy fizjologii ośrodkowego układu nerwowego, a szczególnie procesów neuroprzekaźnictwa. Nadal największe znaczenie w zwalczaniu napadów padaczkowych mają leki hamujące zależne od potencjału kanały sodowe i nasilające przekaźnictwo GABAergiczne, a postępy w tej dziedzinie wiązać można jedynie z łagodniejszą lub wybiórczą modulacją aktywności tych układów. Wydaje się, że receptory jono- i metabotropowe kwasu glutaminowego są wciąż interesującym punktem uchwytu nowych leków przeciwpadaczkowych, jakkolwiek początkowy entuzjazm wokół tych substancji nieco osłabł w świetle ich złożonego mechanizmu działania i możliwości wywoływania objawów niepożądanych.     Streszczenie angielskie: Introduciton. Epilepsy is a common neurological disease but 25-30 p.c. of patients are resistant to pharmacological treatment. Hence the search for new antiepileptics to treat drug-resitant forms of epilepsy and to improve the outcome of recent therapies seems to be fully justified. Objective. The aim of the present paper is a short presentation of new concepts and research directions in pharmacotherpay of epilepsy, with special emphasis on the potential use of neurochemistry and molecular biology achievements. Discussion. Development of ideas on the mechanisms of seizures and antiepileptic drugs reflects the remarkable advancement in the understanding of central nervous system physiology, and neurotransmission in particular. Blockers of voltage-dependent sodium channels and GABA mimetics are still the most important group of anticonvulsants and progress in this field is limited to more selective and more subtle regulations of these neurochemical mechanisms Iono- and metabotropic glutamate receptors are considered to be an attractive target for new anticonvulsants; however, some opinions are now less enthusiastic, since a few antagonists of glutamate receptors display a complex mechanism of action and possess numerous adverse side effects. Despite unquestionable progress in the understanding of the basic mechanisms of epileptogensis, designing antiepileptogenic drugs remains a purely theorectical issue.     Streszczenie angielskie: In order to test if antidepressant drugs can directly influence corticotropin-releasing hormone (CRH) gene expression, their effect on CRH gene promoter activity was evaluated in neuro-2A cells stably transfected with a human CRH - chloramphenicol acetyl-transferase plasmid. Forskolin (an activator of adenylate cyclase), but not phorbol 12-myristate 13-acetate (an activator of protein kinase C), ca. 3-fold increased reporter gene activity, which confirms the critical role of the cAMP-responsive element in regulation of the CRH gene. Imipramine and fluoxetine present in the medium for 5 days, in a concentration-dependent manner (3-30 ćM) inhibited the basal acitivity of CRH gene promoter, while tianeptine was inactive. The obtained results indicate that inhibition of the human CHR gene promoter activity by imipramine and fluoxetine, but not tianeptine, may play a role in mechanism by which the former drugs attenuate HPA axis activity.

    Streszczenie angielskie: It has been hypothesized that some estrogenic metabolites with low hormonal activity, such as estrone or estriol, may serve as endogenous neuroprotective agents. Since estrone naturally occurs in abundance in the human brain, we evaluated its effects on the NMDA-induced toxicity in primary cultures of the rat cortical neurons. The results showed that NMDA (1 mM) had toxic effect on the rat cortical neurons on 12, but not on 7 day in vitro. This effect was partially prevented by DL-2-amino-5-phosphonovaleric acid (AP-5), a competitive NMDA receptor antagonist. Estrone (500 nM) attenuated the NMDA-induced toxicity in estrogen receptor-independent way. These data support the hypothesis that toxic effects of NMDA depend on stage on neuronal development and point to non-genomic neuroprotective actions of estradiol metabolites.

    Streszczenie angielskie: The study examined the effect of some typical and atypical antipsychotic drugs on mouse lymphocyte metabolic and proliferative activity in vitro. The typical antipsychotic drug chlorpromazine (3 x 10**-6, 10**-5 and 10**-4M), significantly inhibited 3H-thymidine incorporation into C57BL/6 mouse spleen cells stimulated by concanavalin A (ConA), lipopolisaccharide (LPS) or pokeweed mitogen (PWM). Chlorpromazine at concentrations of 10**-5 and 10**4 M also suppressed the metabolic activity of splenocytes after ConA stimulation. The atypical antipsychotic agent clozapine (10**-4 and 10**-5 M) decreased the proliferative activity of spenocytes after LPS stimulation, but its inhibitory effect after ConA was observed only at higher concentrations. On the other hand, clozapine did not affect the metabolic activity of splenocytes. Sulpride, a selective dopamine D2 antagonist, at concentrations ranging from 10**-8 to 10**-4 M had no inhibitory effect on the proliferative or metabolic activity of the tested cells. The obtained results indicate that of the three antipsychotic drugs studied, chlorpromazine shows the most potent immunosuppressive effect, clozapine produces a moderate effect and sulpride is totally inactive. These findings suggest that the choice of antipsychotic drugs should also depend on disturbance of immune system activity, in particular those occurring in the several forms of psychosis.
