    Streszczenie angielskie: The present study was undertaken to measure cerebrospinal fluid (CSF) levels of kynurenic acid (KYNA) in patients with relapsing-onset multiple sclerosis (MS) during remission or not progressing for at least 2 months. In these patients the levels of CSF KYNA were found to be significantly lower compared with subjects with non-inflammatory neurological diseases, as well as those with inflammatory disease (median interquartile range): 0,41 (0.3 - 0.5) pmol/ml, n = 26 vs. 0.67 (0.5 - 1.1), n = 23, P 0.01 and 1.7 (1.5 - 2.6), n = 16, P 0.001, respectively). These results provide further evidence of the alterations in the kynurenine pathway during remitting-onset MS.

    Streszczenie angielskie: Lymphoepithelioma is a relatively common malignancy in the nasopharynx. Undifferentiated carcinomas with lymphoid stoma (limhoepithelioma-like carcinomas) of the other organs are very rare. Here we report two cases of such tumours originating in the parotid gland and the uterine cervix. Histologic examination showed nests of undifferentiated epithelial cells (CK+, Vim-) with abundant lymphocytes (predominantly CD+ T cells), plasma cells (Plasma Cell Marker+) and macrophages (CD68+) in and around the tumor cellnests. The Epstein-Barr virus (EBV) genome was detected by in situ hybridization in section from tumour of the parotid gland. The limphoepithelioma-like carcinoma of the salivary gland is often associated with EBV infection. In situ hybridization for the detection of the EBV in malignant epithelial cells was negative in section of uterine cervix.

    Streszczenie angielskie: Activation of latent genes of Epstein-Barr virus (EBV) protects human cells from apoptosis and can induce the expression of bcl-2 gene, a survival factor and inhibitor of apoptosis. We used in situ hybridization technique to detect of EBV LMP1, Bcl-2, Bcl-xL, p21 Cip1 and p53 genes expression in paraffin-embodded tissue from patient with a very rare lumphoepithelial carcinoma (undifferentiated carcinomas with lymphoid stoma) of parotid gland. We detected expression of LMP1, bcl-xL and strong signals from bcl-2. We did not detect signals of p53 and p21 Cip1. We have fixed a potent mechanisms of development of a parotid gland carcinoma: EBV infection or/and LMP1 activation - overexpression of bcl-2 and bxcl-xL - decreased expression of p53 and p21 Cip1 - immortization of cells - cancer.

    Streszczenie angielskie: The cell cycle alternation may cause malignant cell tranformation. The past several years' research has resulted in defining new agents participating in cell proliferation and contributed to the fact that we have started to understand the cell cycle machinery. Molecules involved in basic pathway of cell division are cyclins, the positive regulators of cell cycle, overexpression of which is associated with uncontrolled cell growth and cancer. In our reasearch we examined the level of cyclin genes expression in bone marrow samples from 15 patients with acute leukaemia before therapy. For the detection of cyclin mRNA we used. The Multi-Probe RNase Protection Assay System (Pharmingen). We focused on the expression of D type cyclins. The results we obtained show that leukaemic cells reveal significantly high levels of cyclin D2 and D3 mRNA in majority of examined samples as well as difference in expression of cyclin D1 between ALL and AML cells. The correlation between the clinical data and expression of cyclin genes in hematological malignancies may be a prognostic factor for the relapse or poor prognosis.

    Streszczenie angielskie: B cell chronic lymphocytic leukemia is malignant disorder characterised by accumulation of resting lymphocytes. We investigated the mRNA expression of cyclins A, B, C, D1, D2 and D3, which are characteristic for different phases of the cell cycle, by multi-probe RNase protection assay system. We have examined expression of cyclins in unstimulated and stimulated PMA and LPS lymphocytes in vitro. In our study, all B-CLL tested samples expressed the mRNA cyclin C, D1, and overexpression D2 after stimulation PMA. mRNA cyclin A and D3 appeared in the cases where the cells proliferated in vitro. Induction of cyclin B expression was observed in only 5 cases B-CLL after treating with PMA and LPS.

    Streszczenie angielskie: We analysed biological effect of bergapten and xanthotoxin on human leukaemic cell line - HL-60. The furocoumarins were isolated from a meadow plant - cow parsnip (Heracleum sibiricum L.). To investigate the furocoumarins - mediated effects, the HL-60 cells were incubated with different concentration of bergapten or xanthotoxin (1 ćM, 10 ćM, 50 ćM, 100 ćM, and 500 ćM). Living, apoptotic and necrotic cells were detected by Annexine V method. To monitor the genes expression we used hAPO-2c Template Set of RPA which consist of following Bcl-2 family genes probes: bclw, bcl-XL, bcl-XS, bfl 1, bad, bax, bcl-2, bik, mcl 1, L32, and GAPDH. We established IC50 values - 50 ćM of xanthotin and 100 ćM of bergapten. In our studies decreased expression of bcl-2, bcl-x, bad, bax (Bcl-2 family) and L32 and GAPDH genes (houskeeping) were observed, according to furocoumarins in cultures.
