W wykonywanej w projekcie RAIAP weryfikacji toksyczności pyłów środowiskowych pyły zebrane na różnych stanowiskach europejskich (Oslo, Rzym, Bilthoven, Łódź), wykazujące prozapalne działanie w badaniach in vitro, poddane zostaną badaniom in vivo na szczurach. W pracy zaprezentowano wstępne badania in vivo przeprowadzone w celu validacji i uzyskania danych referencyjnych po dotchawiczej instalacji zawiesiny małych dawek pyłów standardowych PM10.

Streszczenie angielskie: The prevalence of asthma in children and young adults has tripled and quadrupled in many industrialised countries during the last two decades. However, the prevalence of allergic disease in highly industrialized regions contrasts to rates in countries with lower living standards. Environmental influence on the expression of allergic diseases is very likely. In order to verify the toxicity of ambient particulate material which has been identified to show airway inflammation potential in the in vitro screening RAIAP studies, samples collected in different European location (Oslo, Rzym, Bilthoven, Łódź) will be examined in an in vivo animal model. In preliminary in vivo studies performed to validate the system and obtain the refernce data the inflammatory effects were determined using the technique of intratracheal instillation of small samples of resuspended standards PM 10 particulates.

Streszczenie angielskie: The aim of the study was to evaluate the dose-effect and dose-response relationships between the integrated indexes Cd-A ú t (mg/m**3 ú years of exposure) and Cd-B ú years of exposure), and the increase in retinol binding protein excretion in urine (RBP-U) and á2-microglobulin concentration in serum (á2M-S). The study was carried out in the nickel-cadmium battery factory in 1998-1999. Exposure to cadmium was formerly very high. The study group consisted of 116 persons for whom the results of determinations of Cd-B were available during two former observation periods (1983 and 1986-1988). The mean age of the gorup was 49 years and the mean period of exposure was 17 years. The dose-effect relationship between Cd-B ú t and RBP-U or á2M-S was much better (r = 0.642 and 0.513) than between Cd-A út and RBP-U or á2M-S (r = 0.173 and 0.127). There was also correlation between Cd-U (ćg/g creatinine), measured in 1998-1999, and RBP-U or á2M-S (r = 0.343 and 0.198). Urinary cadmium should, however, be used with caution as a dose estimate because its excretion may increase as a result of renal damage. According to the dose-response relationship, an increase in RBP excretion above 300 ćg/g creatinine can be expected in 10 p.c. of subjects at the integrated exposure index (Cd-B ú t) of about 450 ćg/l ú years, and an increase in á2M-S above the accepted cut-off point of 2.4 mg/l can be expected in 10 p.c. of subjects at Cd-B ú t of about 190 ćg/l ú years. The data obtained confirmed the validity of the recommended at present health-based limit for occupational exposure of 5 ćg/l of blood, as well as the superiority of the biological monitoring of exposure to cadmium over the environmental monitoring.
