    Streszczenie angielskie: Tanatopraxis is an intentional action aiming to stop all the processes, which lead to a complete decomposition of a body after death. The words embalment or mummification makes us think about cultures of th eancient Egypt or pre-Columbian America and not about burials of Indo-European nomads or Chinese aristocrats. Meanwhile practices aiming to protect mortal remains against a destructive force of death were known in Europe very long ago. We can see it in paludal burials of the Iron Age and in some testimonies of the ancient epochs. Interventions, which could materially retard decomposition of a body, had been rather generally practiced from the Middle Ages. More practical than religious reasons justified it, but th elatter cannot be overlooked. The religious background of such actions can be seen in the custom of embalming mortal remains of popes or kings of France. It was in Europe where the preservation of corps had started to be treated as a scientific problem, and we can see it for the first time in the Renaissance. Together with a development of medical knowledge and especially of anatomy and preparations, a great progress both in methods an dsubstances used for embalming, took place. Ruysch and Swammerdam in the 17th century, the Hunter brothers in the 18th century and Gannal and Holmes in the 19th century are the milestones in the history of tanatopraxis in Europe. Foundation of the French Institute of Tanatopraxis (IFT) has crowned the many hundred years old experience in the field.

    Streszczenie angielskie: Mechanisms and biochemical consequences of platelet-neutrophil interactions are well known. In contrast, platelet-eosinophil interactions remain largely unexplored. The aim of this study was to assess whether platelets adhere to eosinophils, and to analyze whether selectin-P would mediate that phenomenon. Eosinophils and platelets were obtained from peripheral blood of healthy volunteers. Eosinophils were isolated using magnetic cell separation method. Platelets were isolated and washed. A number of "rosettes" (an eosinophil with more than 5 adherent platelets) per 100 eosinophils were examined in the eosinophil-platelet suspension. Addition of thrombin stimulated formation of "rosettes". Monoclonal antibodies against selectin-P almost completely prevented thrombin-stimulated formation of "rosettes". In summary, intercellular interaction between platelets and eosinophils are mediated by selectin-P. This phenomenon may be of importance in asthma and other atopic diseases.

    Streszczenie polskie: Gracilloplastyka dynamiczna (DGP) jest uznana za skuteczną metodę leczenia schyłkowej fazy nietrzymania stolca (FI), z odsetkiem korzystnych wyników wynoszącym 53-80 proc. W pracy tej opisano nasze wstępne doświadczenia z DGP. Zabieg wykonano u 13 pacjentów z ciężkim nietrzymaniem stolca spowodowanym urazem zwieracza (6), wrodzonym brakiem zwieracza (6) i po resekcji brzuszno-kroczowej (1). DGP przeprowadzono jako zabieg jedno- albo dwuetapowy, zgodnie z klasyczną techniką Baetena. Do stymulacji mięśnia smukłego (m. gracillis) i do konwersji skurczu z szybkiego na tężcowy zastosowano specjalnie przystosowany mikroczipowy generator pulsu IPG 3023 (Medtronic) (MPG). Ocenę przed- i pooperacyjną dokonano w oparciu o skalę Incontinence Scoring System i skalę Holschneidera oraz badania motoryki anorektalnej, ultrasonografię endoanalną i proktografię ewakuacyjna. Jeden pacjent zmarł po pełnej rekonstrukcji anorektalnej z powodu powikłań septycznych wywołanych pooperacyjnym uszkodzeniem odbytu. Nie zaobserwowano żadnych innych poważnych powikłań. Uzyskano zadowalający wzrost spoczynkowego ciśnienia w odbycie i podatności odbytu a także znaczną poprawę w zakresie trzymania stolca. Mimo niewielkiego doświadczenia naszego zespołu, chorobowość i śmiertelność związane z tą metodą leczenia były niewielkie, natomiast wyniki czynnościowe i kliniczne - bardzo dobre. Wstępne doświadczenia ze stosowaniem naszego urządzenia MPG poddawane są obecnie analizie klinicznej. Uzyskano znaczną poprawę jakości życia pacjentów: wszyscy oni w pełni zaakceptowali zastosowane leczenie i wrócili do normalnego życia.

    Streszczenie angielskie: Dynamic gracilloplastry (DGP) has between accepted as an effective treatment for end stage fecal incontinenece (FI), with success rates from 53 to 80 p.c. This paper described our initial experience with DGP. Thirteen patients with severe fecal incontinence due to sphincter injuries (6), congenital absence (6) and one after an abdomino-perineal resection underwent DGP. DHPs were performed as a one- or two stage procedure, acc, to classic Baeten technique. Medtronic IPG 3023 or specially designed microchip pulse generator (MPG) was applied for gracillis stimulation and fast to slow muscle conversion. Pre- and post-operative evaluation was based on Incontinence Scoring System and Holschneider scale as well as anorectal motility studies, endoanal ultrasonography and evaluation proctography. One patient died after total anorectal reconstruction due to septic complications related to postoperative rectal injury. No other serious complications were observed. satisfactory increase of resting anal pressure and rectal compliance as well as marked improvement in anal continenece has been noted. Despite our limited experience morbidity was low while clinical and functional results were very good. Initial experience with MPG are currently under the clinical evaluation. The patients' quality of life significantly improved; all of them fully acepted the applied treatment and returned to normal life.

    Streszczenie angielskie: Recent evidence suggests that hypoxic pulmonary vasoconstriction (HPV) is mediated by hypoxia-induced closure of voltage-gated potassium channels in pulmonary vascular smooth muscle cells. It is also claimed that various vasoconstrictor mediators such as thromboxane A2 (TXA1), platelet activating factor (PAF), cysteinyl leukotrienes (cys-LTs) or endothelin-1 (ET-1) contribute to HPV. Their role, however, has not been unequivocally accepted. On the contrary, it is well known that endothelium-derived nitric oxide negatively modulates HPV. Since NO counteracts action of vasoconstrictor mediators, we tested the hypothesis that modulatory role of TXA2, PAF, cys-LTs and ET-1 in HPV would become apparent in absence of endogenous NO. For that purpose we assessed contribution of these mediators to HPV in the isolated blood-perfused rat lung pretreated with a nonselective NOS inhibitor, L-NAME. HPV, which was greatly augmented by L-NAME (300 ćM) alone, was inhibited neither by a TXA2 synthase inhibitor (Camonagrel, 300 ćM), nor by a PAF receptor antagonist (WEB 2170, 100 ćM), nor by an inhibitor of five-lipoxygenase-activating protein (MK 886, 10 ćM), nor by a non-selective ET-1 receptor antagonist (LU 302872, 30 ćM). In summary, in isolated blood-perfused rat lung, TXA2, PAF, cys-LTs and ET-1 seem not to be involved in HPV, whereas we confirm the dominant role of endogenous NO in blunting HPV.

    Streszczenie angielskie: Eosinophilis accumulation in the airways and sustained eosinophil-derived cysteinyl leukotrienes production repesent key elements of the inflammatory response seen in asthma. However, it is not known whether activated epithelial cells influence cysteinyl leukotrienes production by eosinophils from healthy valunteers. The aim of the present study was therefore to analyse the effects of interactions between non-atopic eosinophils and epithelial cells on cysteinyl leukotrienes production in vitro. We measured cysteinyl leukotrienes released by phorbol 12-myristate 13-acetate (PMA) - activated human eosinophils or epithelial cells (human bronchial epithelial cell line - BEAS-2B) cultured alone or together. While activated BEAS-2B cells barely formed leukotrienes (1.39 pg/ml ń 0.2) (n = 32), activated eosinophilis produced considerable amount of them (62.25 pg/ml ń 10.29) (n = 32). Interestingly, when activated eosinophils and epithelial cells were co-incubated, production of cysteinyl leukotrienes increased substantially (571.1 pg/ml ń 80.9) (n = 32). Thus, eosinophil-epithelial cell interactions, when occur, are associated with increased biosynthesis of cysteinyl leukotrienes.

    Streszczenie angielskie: Induction of NOS-2 in macrophages and smooth muscles within vascular wall with concomittant suppression of endothelial NOS-3 activity is considered to be a hallmark of vascular inflammation that triggers atherogenesis. Accordingly, drugs designed to reverse these changes should not only support vaning function of NOS-3 but also suppress proinflammatory NO production by NOS-2. It means that using selective inhibitors of induction of NOS-2 (they spare ex definitione constitutive activity of NOS-3) is a more rational approach than using "selective" inhibitors of activity to previously induced NOS-2. First of all, those drugs are never sufficiently selective. In our work we tried to identify inhibitors of NOS-2 induction within the group of flavonoids, known stimulators of NOS-3 with putative antiatherogenic effects. Representatives of four main groups of flavonoids: flavonols (kaempferol, quercetin, rutin), flavones (apigenin, primuletin), flavanols (catechine) and flavones (hesperetin, hesperidin, naringenin) were tried on NOS-2 induction and activity in the in vitro model of LPS-treated macrophages (cell line J774.2). While none of these compounds inhibited activity of NOS-2, all with unexpectedly scattered potencies inhibited induction of NOS-2 protein in LPS-treated J774.2 cells, as evidenced by Western blotting technique. Subsequently, RT-PCR and Northern blotting methods revealed that so far the most potent compounds, kaempferol and apigenin, at micromolar concentrations did inhibit NOS-2 induction ...

    Streszczenie angielskie: Nebivolol is a unique á1-adrenoceptor antagonist which possesses peripheral vasodilator properties dependent on endothelial NO. Nebivolol-induced release of NO was attributed to its L stereoisomer and to its ability to stimulate endothelial á2, á3 adrenoceptors or 5 HT1A receptors. Here, in the isolated guinea pig heart we analysed coronary vasodilator potency of L- and D-nebivolol and a possible role of á2, á3 adrenoceptors and 5-HT1A receptors in nebivolol-induced vasodilation. Surprisingly, we ound that not only L-nebivolol (3-30x10**-6 M) but also D-nebivolol (3-30x10**-6 M) induced coronary vasodilation, and both responses were inhibited by L-NAME (10**-4 M). In contrast with the stereoisomers of nebivolol, atenolol at the equimolar concentrations did induce slight casoconstriction. The nonselective á1/á2-adrenoceptor antagonist - nadolol (10-5 M). The selective á3-adrenoceptor antagonist - L 748337 (10**6 M) and the 5 HT1A receptor antagonist - NAN 190 (5 x 10**-6 M), none of them inhibited coronary vasodilation induced by D- and L-nebivolol. In summary, in the isolated guinea pig heart both D- and L-nebivolol act as coronary vasodilators. Coronary vasodilation induced by stereoisomers of nebivolol is mediated by endothelium-derived NO and does not depend on á2, á3 adrenoceptors or 5 HT1A receptors.

    Streszczenie angielskie: It has been observed that ticlopidine and clopidogrel show, apart from their delayed antiplatelet properties, an immediate and transient thombolytic action related to the ability of these thienopyridines to stimulate the secretory function of vascular endothelium. With the objective to construct new molecules with identical thrombolytic potency but at a higher level, we carried out different structural modifications in the thienopyridine chemical molecule to conclude that the presence of a second N atom in the pyridine cycle (yielding pyrimidine moiety) and the presence of an additional cycle fused to the thienyl ring would lead to enhanced thrombolytic effects. Here we report the six-step synthesis of a series of new benzothienopyrimidinone derivatives characterized by this searched for potent thrombolytic activity. The pharmacological assay used anaesthetised Wistar rats with extracorporal circulation in which arterial blood superfused thrombi adhering to a strip of collagen. Weight of thrombi was continously monitored. Six compounds of the series were much more potent thrombolytic agents than their thienopyridine references: the effective thrombolytic dose that produced 30 p.c. of maximum thrombolysis (ED 30) was at a range of 8 to 170 ćg kg**-1 as compared with ED30 values of 16000 to 20000 ćg kg**-1 for clopidogrel and ticlopidine respectively.     Streszczenie angielskie: Prostaglandin endoperoxide H2 (PGH2) is generated from arachidonic acid by either consitutive (COX)-1) or inducible (COX-2) cyclooxygenases. In arterial wall PGH2 is converted by PGI2 synthase (PGI-S) to prostacyclin (PGI2), and in plateles by thromboxane synthase (TX-S) to thromboxane (TXA2). Other prostanoids as PGD2, PGF2ŕ or PGE2 were belived to arise non-enzymatically from PGH2. Only recently , human prostaglandin E synthase (PGE-S) has been identified and cloned as a membrane bound, microsomal, glutathione-dependent inducible enzyme. Here we demonstrated that interleukin 1 á (IL-1á) is an inducer of COX-2 and PGE-S in human umbilical vein endothelial cells (HUVEC). Functional expression of PGE-S was measured at the level of specific mRNA by semi-quantitative RT-PCR, PGE-S protein was detected by Western blot in HUVEC, while PGE2 was measured by immunoassay in the supernatant. Actinomycin D, a classical transcription inhibitor, was used to prove that indeed IL-1á induced the functional PGE-S enzyme. PGE2 generation in HUVEC was inhibited by indomethacin, acetamoniphen and dexamethasone. In conclusion, we found that in cultured endothelial cells IL-1á induced as evidenced by the appearance of its transcript and its functional enzyme. The induction of endothelial PGE-S and COX-2 appeared to be and their transcripts were induced as fast as one might expect from immediate early genes. It means that IL-1á-triggered-PGE2 biosynthesis in endothelial cells is probably regulated by induction of both COX-2 and ...

    Streszczenie angielskie: Increased generation of reactive oxygen species contribute to endothelial dysfunction in atherosclerosis, hypertension and heart failure. Recently, it was suggested that bursts of superoxide anions may inactivate endothelial surface-bound enzymes such as angiotensin converting enzyme (ACE). Here, we tested effects of xanthine/xanthine oxidase-derived superoxide anions on vascular responses and ACE activity in the isolated guinea pig heart. We anlysed effects of intracoronary infusion of low concentration of xanthine oxidase (10 mU/ml) in the presence of xanthine (0,5 mM) (X/XO) on bradykinin, other endothelium-dependent and independent vasodilators (acetylcholine, ADP, SNAP), as well as vasoconstrictor responses to angiotensin I and angiotensin II. Surprisingly, X/XO significantly augumented coronary response to bradykinin without an effect on response to ADP, acetylcholine, SNAP, angiotensin I and angiotensin II. In contrast, inhibition of ACE by perindoprilate (100 nM) resulted in augumentation of bradykinin-induced vasodialidation as well as diminution of angiotensin I-evoked vasoconstriction without an influence on other responses. In summary, in the isolated guinea pig heart, X/XO-derived free radicals selectivity augumented coronary vasodilator response to bradykinin, which cannot be explained by X/XO-induced derangement of ACE.
