Abnormal proteolytic degradation of the á amyloid precursor protein (á-APP) may result in accumulation of potentially neurotix á amyloid (áA). The role of various receptors in the regulation of á-APP processing has been suggested. This study aimed to determine how NMDA receptors and Caý+ ions regulate proteolysis of á-APP in rat hippocampus in vitro. Adult rat hippocampal slices were superfused with NMDA-containing media, and immunoreactivity of soluble á-APP derivatives was detected in dialysates. Application of 100 ćM and 250 ćM NMDA for 20 min in Caý+-containing medium induced dose-dependent release of amino-terminal á-APP derivatives, and a fragment of Aá sequence, whereas carboxy-terminal fragments of á-APP were only slighty detected. This indicates activation of á-APP processing, and release of its soluble cleavage products. This effect was inhibited by NMDA receptor antagonist 1 ćM MK-801 and 100 ćM CPP in Caý+-free medium, thus indicating that NMDA receptors and calcium ions mediate proteolytic non-amyloidogenic degradation of the á-APP.

Previously, a large two-part inter-laboratory round robin endotoxin assay study was completed. This first study showed that when cotton dust samples, which are practically identical, are assayed for endotoxin that the intra-laboratory results had a very small variation while intra-laboratory results of the sample had a very high variation. In the first part of the study, each laboratory results of the sample had a very high variation. In the first part of the study, each laboratory followed its own in-house assay protocol; but in the second part of the study, when the extraction protocol was standardized, the inter-laboratory results showed a lower variation, which suggested that with further standardization, further reduction of differences between laboratories might be achieved in order that results between laboratories would become more comparable. The results stimulated interest in extending the study to include cotton dust with two levels of endotoxin, standardization of the extraction proteocol, and using the same assay kit from the same production lot. The results of this second round robin endotoxin assay study indicate that differences between laboratories are still high, but most of the laboratories could discern the cotton dusts with the different levels of endotoxin.

The aim of this study was to examine the prevalence of permanent teeth enamel anomalies in schoolchildren in one warsaw schools. There were 409 children examined (7-14 years old), living from the birth in a region with optimal level of fluoride in the water. The epidemiological DDE Index was used in this examinations. 38 p.c. of children had enamel anomalies. The most of examined children had disturbances in from of white/cream mottling and lines. The localization was referred to premolar teeth then incisors and canines in maxilla.
