    Streszczenie angielskie: Spatial and temporal relations between metalloproteinase (MMP-2 and MMP-9) activation and laminin degradation in garbil hippocampus after transient cerebral ischemia has been studied. Activity of MMPs was determined by gelatin zymography in homogenates from dorsal (DP, an equivalent of CA1 sector) and abdominal (AbP, containing CA2-4 and gyrus dentatus) parts of hippocampus. A significant activation of both investigated metalloproteinases was found at 72 h of recovery. Whereas MMP-2 up-regulation did not show any spatial preferences, the increase of MMP-9 activity was observed exclusively in DP. Activation of MMP-9 at this time point correlated spatially with degradation of laminin - protein of extracellular matrix. These results show that MMP pathway may function as a component of delayed neuronal death cascade in the apoptogenic CA1 sector after transient global ischemia.

    Streszczenie polskie: Istnieje rosnące zainteresowanie komórkami macierzystymi jako potencjalnymi "narzędziami" w przyszłej terapii wielu chorób, w tym również neurologicznych, w których mogłyby one zastąpić uszkodzone nieodwracalnie komórki, a nawet narządy. Ze względu na nierozwiązane problemy etyczno/prawne wiążące się z użyciem płodów, a nawet wytwarzaniem zarodków ludzkich, zainteresowanie badaczy zwróciło się w kierunku tzw. "dojrzałych", tkankowo- specyficznych komórek macierzystych znajduwanych w dojrzałych narządach. Badania ostatnich lat wykazały, że komórki zwierzęce (a w ostatnim roku potwierdzono to również u ludzi) posiadają niezwykłe cechy plastyczności, znacznie przekraczające ramy klasycznej embriologii i z powodzeniem mogą być stosowane jako źródła samo-odtwarzających się, ukierunkowanych komórek prekursorowych typowych nie tylko dla wyjściowej, ale i innych tklanek. W artykule krótko omówiono następujące zagadnienia: Jakie są cechy charakterystyczne różnych rodzajów komórek macierzystych i gdzie można je znaleźć; Jakie dowody przemawiają za skutecznością użycia komórek macierzystych w terapii OUN; Jakie są najważniejsze problemy do rozwiązania przed zastosowaniem komórek macierzystych w klinice. Ocena korzyści i ryzyka; Proponowane strategie i cele terapeutyczne w leczeniu schorzeń OUN.

    Streszczenie angielskie: There is a growing interest in medical potential of stem cells which may be used someday to create new tissue. Sugested applications involve a board spectrum ranging from the replacement for cells destroyed by diseases to even organ transplants. As ethical and legal controversy makes uncertain the future of embryonic stem cells research, attention has been recently turned to adult stem cells to discover whether they also can serve to transplantation. In this article we present data indicating that adult stem cells found in areas of the body like blood, skin, lymph and nervous systems, may be more versatile than previously assumed and in certain conditions can broke tissue barriers for differentiation. Thus, in this respect they can behave like their own pluripotent ancestors, although underlying mechanisms for this phenomena are still not clear. Furthermore, the following issues are shortly discussed: What are the characteristics of the different stem cells and where they can be found; What are experimental evidences that stem cells can be used for brain repair; What are the main problems must be solved before their clinical application and the risk- benefit assessment; The possible strategies and targets for stem cell therapies of neurological diseases.
