    Streszczenie angielskie: The cells with nuclear DNA fragmentation related to apoptosis were detected by TUNEL technique in the seminiferous epithelium of control rats and of rats with experimental hyperprolactinemia induced by metoclopramide. The percentage of convoluted tubules with apoptotic cells and the number of apoptotic cells (predominantly spermatogonia and spermatocytes) was increased in the experimental group. The results indicated stage-specific germ cell apoptosis. In the experimental group, apoptotic cells were most evident at early (I-IX), middle (VII-VIII) and late (XII-XIV) stages of teh seminiferous epithelium cycle, as revealed by light and electron microscopy. We suggest that a decreased concentration of testosterone and an increased concentration of prolactin could disturb spermatogenesis and contribute to the intensive apoptosis of germ cell in rats with hyperprolactinemia. sertoli cells which have receptors for testosterone and prolactin and play an important role in spermatogenesis and in the initiation of apoptosis in seminiferous epithelium, could mediate such an influence of both hormones.

    Streszczenie angielskie: The aim of the study was to standardize a dependable method, which would be used to differentiate thyroid carcinoma in fine-needle biopsies and to adapt radiofrequency ablation techniques to safety treat benign thyroid tumors. Fine-needle aspiration biopsies, taken pre- and intraoperatively were assessed cytologically and histologically. Immunochemical markers (PCNA, Ki67 and AgNOR) were used to differentiate between carcinoma and benign folicular adenomas. Apoptosis was studied in benign tumors using TUNEL, Bcl-2, CD95 and Bax immunoassays. Ablation was performed intraoperatively on thyroid tissue fragments ranging in size from 1,3 to 2,2 cm, mean 1,7 ń 0,5 prepared for resection with full blood perfusion of the gland. AgNOR silver staining was used to assess the number of active nucleoi "NORs". Benign proliferation showed a number of 1-2 nuceloli, in adenomas 4-6 nucleoli and 8 nucleoli in malignancies. PCNA showed highest specificity where 80 p.c. of cancerous cells had positive results. The Ki67 marker in primary cancer showed no staining and stained positive in 50 p.c. of metastatic cancer. Apoptosis was also not increased in the case of adenomas. In the 58 patients undergoing the procedure we noted in 4 patients (6,1 p.c.) temporary laryngeal nerve paralysis, in 2 (3 p.c.) cases minute bleeding after needle arm retraction. To conclude: 1) the suggested assay panel is able to discriminate between follicular carcinoma and adenoma, 2) radiofrequency ablation can be an alternative to surgical methods in neutral nodular goiter, 3) necrosis zone of the ablated thyroid lesion is closely correlated to the used ablation parameters.
