    Streszczenie angielskie: Caspases are crucial mediators of apoptosis, a form of physiological cell death. Their activation is carefully controlled by a philogenetically conserved death program, which is indispensable for the homeostasis and development of higher organisms. Dysregulation of apoptosis contributes to the pathogenesis of many human diseases. As effectors of the apoptotic machinery, caspases are considered potential therapeutic targets. In vitro studies have demonstrated the requirement of caspase activity for both the triggering phase as well as the execution of apoptosis, thus providing a molecular base for the fine-tuning of this process by pharmacological agents. The precise roles of the individual caspases in vivo and their functional relation to each other have been best demonstrated in genitically modified animals. The generation of single caspase-deficient mice have confirmed most of the data obtained in vitro and exposed some new aspects previously undetected in teh cell culture system. Interestingly, inactivation of many caspases revealed not only their expected participation in apoptotic as well as in the maturation of cytokines, but also provided hints about the role of at least some caspases in cell differentiation and stimulatory responses. In this review we will discuss what these studies have unveiled about the role of individual caspases in development, apoptosis, and inflammation, with particular focus on their role beyond the apoptotic process.

    Streszczenie angielskie: Background: The objeactive was to compare the clinical features of early local Lyme Disease: Erythema Migrans (EM) baeatween two highly endemic areas: New York State and Northeastern Poland. Study was based on self reporting questionnaires, physical examination, and laboratory results. Material/Methods: Group I (n = 27) consissted of citizens from New York State presenting with EM, referred to the study by their physicians. Group II (n = 25) consisted of patients treated as outpatients in 1998 - 1999 at the Department of Infectious Diseases and Neuroinfections, Medical University of Białystok, Poland. The diagnosis of EM was based on patient history and clinical picture. Results: Depending on the immune reaction of the host, the clinical picture is variabale. The initial symptoms range from discrete to strongly expressedd. The inflammatory reaction at the site of tick-bite seems to be milder in Europe than in North America, which is reflected in teh prevalence of the annular form of rash and milder expression of general symptoms. Mildor absent general symptoms and the prevalence of the annular over homogenous form are observed in those areas of Europe where Ixodes ricinus is the main veactor. In the endemic area of New York State, where Ixodes scapularis mainly occurs, uniform EM with fluplike symptoms is more common. Conclusions: There are distinct differences in the typical clinical picures between the two separated endemic areas of New York State and Northeastern Poland, due to different vectors (salive components?), bacaterial genospecies, likely intermediate hosts and clinatic differences. Familiarity with the presenting form of the disease can be very important, especially for primary care physicians.
