    Streszczenie angielskie: There are 9 channels of the CIC family in mammals and new others in fishes, plants, yeast and bacteria. The CIC channels are present in different tissues and play a role in transmembrane potential stabilization, transepithelial transport, cell volume regulation, acidification of intracellular organelles. The genetic defects of CIC-1 chloride channel lead to myotonias, the defect in CIC-5 channel to the formation of stones in kidney, while the defect in CIC-Kb channel leads to the Bartter's syndrome.

    Streszczenie angielskie: Background. A decrease in mortality after myocardial infarction (MI) may be achieved by the improvement in risk stratification and implementation of more effective therapy. Heart rate (HR) calculated from a standard 12-lead ECG in a simple and widely accessible parameter which may be used for the assessment of prognosis after MI. Aim. To assess the prognostic value of HR derived from a 12-lead ECG recorded after the acute phase of MI and to formulate a prognostic index consisting of simple clinical and ECG parameters. Methods. The study group consisted of 309 consecutive patients (95 females, mean age 61.7 ń 12.8 years), who survived the first 48 hours of acute MI. HR derived from a 12-lead ECG recorded between 3 and 5 days after the onset of MI and other parameters were used to assess the risk of cardiac death during the one-year follow-up period. Results. During one year of follow-up, 72 (23.3 p.c.) patients died dut to cardac causes. The mean HR was significantly higher in patients who died than in survivors (89.8 ń 29.5 versus 66.2 ń 12 beats/min, p 0.0001). The multivariate analysis revealed that HR, age, Kilip class and contraindications to beta blockers were the independent parameters of prognostic significance. The prognostic index consisting of HR 75 beats/min, age 60 years, Kilip class 1 and lack of beta-blocker therapy identified patients at risk of cardiac death with a sensitivity of 75 p.c. and specificity of 100 p.c. Conclusons. HR derived from a standard ECG recorded outside the acute phase of MI is higher in patients at risk of cardiac death after MI than in survivors. HR and other clinical as well as demographic parameters provide a simple prognostic index after MI.

    Streszczenie angielskie: The glycine receptor belongs to the ligand-gated ion channel superfamily. It is a chloride conducting channel composed of four transmembrane domains. It was previously shown that the second transmembrane domain (M2) of the glycine receptor forms an ion conduction pathway throught lipid bilayers. The amino-acid sequence of the transmembrane segment M2 of the glycine receptor has a high homology to all receptors of the ligand-gated ion channel superfamily. In our report, we have used a synthetic M2 peptide. It was incorporated into a planar membrane of known lipid composition and currents induced by M2 were measured by the Black Lipid Membrane technique. When the planar lipid bilayer was composed of 75 p.c. phosphatidylethanolamine and 25 p.c. phosphatidylserine, the reversal potential measured in a 150/600 mM KCl (sic/trans) gradient was - 19 mV suggesting that the examined pore was preferential to anions, PK/PCl = 0.25. In contrast, when 75 p.c. phosphatidylserine and 25 p.c. phosphatidylethanolamine was used, the reversal potential was +20 mV and the pore was preferential to cations, PK/PCCl = 4.36. SIngle-channel currents were recorded with two predominant amplitudes corresponding to the main-conductance and sub-conductance states. Both conductance states (about 12 pS and 30 pS) were measured in a symmetric solution of 50 mM KCl. The observed single-channel properties suggest that the selectivity and conductance of the pore formed by the M2 peptide of the glycine receptor depend on the lipid composition of the planar bilayer.
