Streszczenie angielskie: It has been found that trypsin and ŕ-chymotrypsin inhibitors isolated from Ascaris suum act embryotoxically and teratogenically on White Leghorn chicken embryos. Mortality rate for the chicken embryos on day 15 of incubation was 45.0 ń 3.5 p.c. after injection of trypsin inhibitor and 44.0 ń 3.5 p.c. after adminstration ŕ-chymotrypsin inhibitor. Gross examination of surviving embryos and their dissection revealed pathological changes (abdominal dropsy, umbilical hernia, subcutaneous oedema, hemoperitoneum, hemopericardium), symptoms indicating retardation in growth (lack of down, retarded ossification of long bones, decreased mean body weight) as well as malformations (schistocelia, micrognathia, cyclopia, crossed beak, cranial deformities) after injection of inhibitors from Ascaris. The highest incidence of embryos with pathological changes and malformationsd was foundf after administration of ŕ-chymotrypsin inhibitor. The most commonly occurring abnormality was schistocelia (21.4 ń 3.88 p.c.). Growth malformations were not found in the control groups. The trypsin and ŕ-chymotrypsin inhibitors present in Ascaris homogenate have a significant disturbing effect on ther development of the chicken embryo.

Streszczenie angielskie: Ontogeny of the catecholaminergic system of the preoptic area (PA) was studied in various animal species including mice, rats, cats and lower vertebrates. Until now, there has been no data about development of catecholaminergic structures in the porcine PA. To study this problem, hypothalami from six groups of animals were collected. Three groups of foetuses (70, 84 and 112 days old) and three groups of female pigs (1 day, 10 weeks and 7 - 8 months old) were used. Nerve structures immunoreactive for the studied substances: tyrosine hydroxylase (TH), dopamine á-hydroxylase (DáH) and phenylethanoloamine-N-metylthransfrease (PNMT) were observed in different periods. In PA, TH-IR (immunoreactive) structures appeared before 70th day of foetal life, DáH-IR between 70th and 84th day of foetal live and PNMT-IR only in 10-week old and adult animals. In the PA of 70-day old foetuses, single smooth and varicose nerve fibres immunoreactive only to TH were found. In PA of 84-day old foetuses, additionally, single nerve cell bodies immunoreactive to TH were shown and some of them also contained immunoreactivity to DáH. In Pa of 1-day old piglets, moderate numbers of nerve fibres immunoreactive to TH and only single TH/DáH-IR nerve terminals were observed. TH-IR nerve cell bodies were also moderate in number and many of them contained simultaneously immunoreactivity to DáH. In PA of 10-week old pigs, a moderate number of immunopositive nerve fibres was observed. They contained mainly TH, but part of them stained also for TH/DáH. Only very few nerve fibres containing exclusively DáH were found...

Streszczenie angielskie: The aim of the present study was the evaluation of the expression levels of PDGF-A and B, and PDGFR-ŕ, and changes of apillary cross-sectional areas during terminal vascular network formation in the developing lung. Pathogen-free fetuses and newborn Wistar rats were used. Lungs were processed for frozen sections used for immunohistochemical staining and parallel tissue specimens were prepared for transmission electron microscopy. The expression levels of PDGF-A and B, and PDGRR-ŕ were obtained using quantitative image anlysis. Additionally, the morphometry of capillary cross-sectional area was performed. We found that the expression levels of the studied antigens fluctuated, and the highest values were found in 2-day old fetuses (p 0.0001). Although there were no significant differences of the area of "a single" capillary, the area of the endothelium decreased significantly in perinatal period (p 0.0001). We conclude that the statistically significant changes of the studied antigens' expression as well as the of capillary cross-sectional area occur in the perinatal period.

Streszczenie angielskie: Development of thymic medulla was examined on consecutive gestational days (GD) in Wistar rats. Medullary thymic epithelial cells (TEC) were identified by immunocytochemical localisation of neuronspecific enolase (NSE). Organisation of thymic medullary architecture was determined by interaction of thymocytes with NSE-positive TEC, that led to formation of lymphoepithelial complexes (GD 19), in which the cells exhibited proliferative activity or traits of apoptosis. The studies indicated that differentiation events and organisation of thymic medulla require stagae-specific interactions between TEC and thymocytes.

Streszczenie angielskie: According to epidemiological studies, it is possible that some secondary metabolites of indoor airborne fungi could be responsible for health troubles which occupants sufer from. In our previous experiments, a model with tracheal rings of 1-day-old chicks in vitro was shown to be a very suitable method to study the ciliostatic chloroform-extractable endo- end/or exometabolites of filamentous fungi. In this study we isolated the filamentous fungi from walls of "moudy" dwellings and schools (cultivation on dichloran 18 p.c. glycaerol agar at 25 and 37řC for 10 d) in Slovakia. We studied the ciliostatic effect of the chloroform-extractable endo- and exometabolites of 96 representative isolates (Stationary cultivation on the liquid medium with 2 p.c. of yeast extract and 10 p.c. of sucrose at 25řC for 10 days) on the cilia movement in tracheal organ cultures of 1-day-old chickens in vitro after 24, 48 and 72 hrs (incubation in the minimal essential medium according to Eagle with Earl's salts and 20 ćg of extract of metabolites dissolved in dimethylsulfoxide per 1 mL). Strains of Penicillium Link: Fr. sp., Aspregillus versicolor (Vuill.). Tiraboschi, A. flavus Link, Cladosporium sphaerospermum Penzig adn C. cladosporioides (Fres.) de Vries were isolated most frequently. Two A. flavus isolates were able to produce aflatoxins B1, B2, G1, G2 in vitro after cultivation on the liquid medium with 20 p.c. sucrose and 2 p.c. yeast extract. This is the first isolation of aflatoxigenic A. flavus strains from dwellings in Slovakia. All frequently isolated strainsproduced secondary metabolites...

Streszczenie angielskie: It has been hypothesized that some estrogenic metabolites with low hormonal activity, such as estrone or estriol, may serve as endogenous neuroprotective agents. Since estrone naturally occurs in abundance in the human brain, we evaluated its effects on the NMDA-induced toxicity in primary cultures of the rat cortical neurons. The results showed that NMDA (1 mM) had toxic effect on the rat cortical neurons on 12, but not on 7 day in vitro. This effect was partially prevented by DL-2-amino-5-phosphonovaleric acid (AP-5), a competitive NMDA receptor antagonist. Estrone (500 nM) attenuated the NMDA-induced toxicity in estrogen receptor-independent way. These data support the hypothesis that toxic effects of NMDA depend on stage on neuronal development and point to non-genomic neuroprotective actions of estradiol metabolites.

Streszczenie angielskie: The authors present the animal model for open fetal surgery as a first step in preparing human open fetal surgery. The second purpose of the study was investigation of fetal cutaneous wound healing. Six time-dated pregnant ewes (110-120 day's gestation) underwent general anesthesia. Then after laparotomy, histerotomy was performed to prepare diaphragmatic hernia in the fetus. During the second histerotomy surgical practise in repair of diaphragmatic hernia was performed. This paper deals with the problems of monitoring fetuses during operation, preventing premature labour and methods of opening and closing the womb. Specimens of healing tissues were histologicaly investigated to compare the manner of fetal and adult wound repair.
