    Streszczenie angielskie: The present study was designed to investigate whether quercetin, a very common flavonoid widely distributed in many plants, can induce apoptosis in monkey kidney cells (GMK). Involvement of the expression of heat shock proteins Hsp72, Hsp3, Hsp27 and the significance of cell culture model in this process were also examined. The studies have shown that quercetin alone and in combination with the heat shock can induce apoptosis and necrosis in vitro in the sutided cells, but the percentagae of affected cells did not exceed 3.9 p.c. In the same experimental conditions, the expression of Hsp73, Hsp72 and Hsp27 increased in cells cultured in two-dimensional systm and decreased in three-dimensional model. This indicates that strong inhibition of heat shock proteins in GMK cells is not correlated with an adequate increase in the sensitivity of cells to undergo apoptosis. It also shows that the sensitivity of cells, manifested by Hsp expression and apoptosis induction, depends on the culture model and culture conditions.

    Streszczenie angielskie: Adriamycin induced DNA damage and repair were investigated by the comet assay in four human cell lines (CRL2088, ME18, Lu106, WISH) and five animal cell lines (L929, Balb/3T3, CHO, MDBK, Vero). The results indicated the concentration-dependent induction of DNA strand breaks in all cell lines after adriamycin treatment. Simultaneously, wide differences in sensitivity of cell to the damaging action of adriamycin were observed. The most sensitive were two human cell lines derived from epithelial tissues, Lu106 and WISH. In these lines the breaks induced by adriamycin were not repaired within 6 h postincubation. In two other human cell lines, CRL2088 and ME18, only a small number of DNA breaks appeared after adriamycin exposure and these were very quickly repaired. Among the animal cell lines only the mouse cell line L929 was very sensitive to adriamycin, but most of the induced breaks were repaired within 6 h postincubation. The remaining animal cell lines were less sensitive to DNA damage by adriamycin and all except 3T3, completely repaired the breaks within 6 h postincubation.

    Streszczenie angielskie: Xenotransplantation represents a promising solution to the ever increasing shortage of donor organs in allotransplantation. However, due to different and stronger models of rejection, successful xenotransplantation will require different organ-protective regimes from those used in allogenic transplantation today. Since one can simply increase the dosage of the drugs used, immunomodulation or tolerance induction of the recipient would be the most desirable approach. Transfusion of donor leukocytes has been shown to downregulate recipient responses or even induce peripheral tolerance in small animal models. Since the infusion of donor cells represents a relatively simple approach, as one can purify and compose the inoculum exactly before infusion, we studied whether this approach can be successfully employed in a preclinical swine to nonhuman primate model of peripheral tolerance induction/immunomodulation. In our model, baboons underwent sequential column adsorption and complement blockade. The animals received only initial immunosuppression with cyclophosphamide. No further immunosuppression was given. Subsequently all animals received 1-3x10**10 porcine splenocytes i.v. Development and maintainance of chimerism was analyzed by sequential flow cytometric and PCR analyses. Other parameters studied included effects of the preparatory induction protocol. We could show that a low level of chimerism is maintained in these animals for up to 1,5 years, despite the fact that they received no additional immunosuppression after the initial one.     Streszczenie angielskie: An investigation was conducted into the differences between macrosmatic and microsmatic animals if arterial supply in the area of the olfactory trigone and the anterior perforated substance. A brain of domestic cat was taken as an example of a macrosmatic animal and a brain of baboon as an example of a micorsmatic animal. The cerebral blood vessels of the cadavers of 30 cats and 11 baboons were filled with latex and, after fixation in acid alcohol by means of microscopic operation, the cortical and deep branches of the anterior and middle cerebral artieres were dissected. We discovered that there were differences between these two groups of animals in the places of arising of deep branches and in their course. In the cat the deep branches running through the area of the olfactory trigone arose from the beginning part of the middle cerebral artery in numbers 5 - 10 and entered the brain in the olfactory tubercle. In the baboon the deep branches arose from two sources: the middle cerebral artery and the anterior cerebral artery. The branches of the middle cerebral artery went through the anterior perforated substance, forming two groups of branches: lateral and medial.

    Streszczenie angielskie: Background: The effects of long-term estrogen (E2) replacement therapy (ERT) on circulating and bone marrow total plasma nitric oxide (NOx) levels were determined in rhesus monkeys. In addition changes in circulating NOx during the menstrual cycle were measured. We measured Nox levels in 20 ovariectomized animals and 27 cycling animals. In the first group monkeys were ovariectomized (OVX; n=10) and treated with estrogen implants for 21 months. The control group (n=10) was OVX and received vehicle implants only. After 21 months systemic NOx status was measured. In a second group of cycling animals NOx was measured during the follicular and luteal phases, and correlated whit body comparison parameters determined by dual energy X-ray absorptiometry (DXA) analysis. Results: Replacement of estrogen in OVX monkeys resulted in reduced circulating but not bone marrow plasma NOx compared to OVX monkeys. Circulating NOx levels in these animals correlated negatively (rý= -0.35; P=0.03) with estradiol levels. In cycling monkeys luteal levels of NOx were found to be significantly higher than follicular levels and correlated negatively (rý=-0.4; P=0.036) with estradiol but not progesterone levels. Negative correlations between NOx levels and body weight, fat and lean mass were also found. Conclusion: Long-term ERT influences systemic NOx levels in negative fashion without effects on bone marrow plasma NOx. Circulating NOx was higher during the luteal phase.
