    Streszczenie angielskie: Blood from the glandular part of hamster gastric mucosa is drained by collecting venules, running from the subepithelial layer towards lamina muscularis mucosae. To visualise vessels involved in the venous outflow, hamsters were exposed to atropine and subjected to intravital ligation of portal vein, causing strong hyperaemia. Distribution of vessels and their connections were studied a) in translucent, flat preparations of the glandular stomach, b) in thic sections of glandular mucosa cleared in the mineral oil, and c) in semi-thin plastic or parafin sections. Collecting venules were drained by single vessels running parallel to the lamina muscularis mucosae (paramuscular venules), which, in turn, joined submucosal veins through opening in muscularis mucosae. Moreover, some collecting and paramuscular venules discharged into venules belonging to vascular triples composed of two venules and arteriole. The triplets were also parallel to muscularis mucosae. Triplets did not form connections with submucosal veins but passed on the abluminal surface of muscularis mucosae. Thus, some structural elements involved in venous outflow from glandular gastric mucosa differ from those in rats, in which vascular triplets were absent and all collecting venules drained into single paramuscular vessels. Conctraction/relaxation of muscularis mucosae may regulat the amount of blood present in the venous system of the mucosa and the diameter of venules. Rhythmical changes of the latter could cause changes in intramucosal pressure, affecting movement of tissue fluid in the mucosa and thus the functio of gastric cells.

    Streszczenie angielskie: Specificity of targeting of the oxidative stress towards lipid and protein fractions in a model of estrogen-induced Syrian hamster neprhocarcinogenesis was evaluated. The amount of proteins modified by oxidative stress was significantly elevated as early as one month after the initial implantation of estradiol to the experimental versus the control group, while the stress did not afect lipids. Subcellular localization of the oxidative stress target wass determined by the analysis of protein oxidation in subcellular fractions of kidney cells. The endoplasmic reticulum membranes were the fraction most affected bay the oxidative stress.

    Streszczenie angielskie: Carcinoembryonic antigen (CEA) is an oncofoetal cell surface glycoprotein that serves as an important tumour marker for colorectal and some other carcinomas. Its immunoglobulin-like structure places CEA within the immunoglobulin superfamily. CEA functions in several biological roles including homotypic and heterotypic (with other CEA familiy members) cell adhesion. Cell-cell interaction can be modulated by different factors, e.g., post-transplational modifications such as glycosylation. The purpose of this study was to examine whether changes in carbohydrate composition of CEA oligosaccharides can influence homotypic (CEA-CEA) interactions. In order to modulate glycosylation of CEA we used two different glycosylation mutants of Chinese hamster ovary (CH)) cells, Lec2 and Lec8. Lec2 cells showuld produce CEA with nonsialylated N-glycans, while Lec8 cells should yield more truncated sugar structures than Lec2. Parental CHO (Pro5) cells and the glycosylation deficient mutants were stably transfected wtih CEA cDNA. All three CEA glycoforms, tested in a solid-phase cell adhesion assay, showed an ability to mediate CEA-dependent cell adhesion, and no qualitative differences in the adhesion between the glycoforms were observed. Thus, it may be assumed that carbohydrates do not plas a role in homotypic adhesion, and the interactions between CEA molecules depend solely on the polypeptide structure.

    Streszczenie angielskie: Classical and ultrastructural neuropathology of prion diseases are generally well described. Here we report that alternations of myelinated fibres in hamsters infected either with polioencephalopathic strains of scrapie or panencephalopathic strains of CJD (Echigo-1) are virtually identical and differ only quantitatively. In contrast, mice infected with the panencephalopathic Fujisaki strain of CJD exhibited much more elaborate changes of myelinated fibres.

    Streszczenie angielskie: We report hare the microglial (macrophage) and astrocytic reaction in several models of transmissible spongiform encephalopathies (TSEs) or prion diseases. With the low power electron microscopy it was readily apparent that myelinated vacuoles were surrounded by cells and their processes. The latter belonged either to hyperplastic reactive astrocytes or to macrophages. Typically, reactive astrocytes exhibited cytoplasm filled with innumerable glial filaments and, occasionally, other organelles (like cilia) and abundant tortuous intercellular junctions of adhesive plaque junction type. Desmosome-like junctions connecting astrocytic elements were also seen. As described earlier, astrocytic process were occasionally interdigitated with oligodendroglial cells and their processes. Two types of macrophages were readily described. The majority of them exhibited electron-dense cytoplasm and numerous "empty" vacuoles (digestive chambers) containing cellular debris. Occasional vacuoles were surrounded by a thin collar reminiscent of "lyre-like inclusions" of the second type of macrophages. Several mylinated fibres were clearly engulfed by the cytoplasm of a macrophage containing unusual annulate lamellae.

    Streszczenie angielskie: As in other neurodegenerative diseases such as Alzheimer's disease, neurons in prion diseases or transmisible spongiform encephalopathies (TSEs) die via programmed cell death of which the apoptotic process is relatively well characterized. A subcellular alteration linked to apoptosis is the formation of autophagic vacuoles, which we and others demonstrated in CJD- and scrapie-affected rodent brains. Autophagy may co-exist with apoptosis or may precede it and the process may be induced by apoptosis stimuli. Here, we extend these observations using different model of scrapie and CJD. Both scrapie models (the 265K and 22C-H) demonstrated autophagic vacuoles with the same frequency; hence, they will be described together. While the following changes had been observed simultaneously in different areas of the same sample, this description is organised as if it followed a sequence of events. First, a part of the neuronal cytoplasm was sequestrated by concentric arrays of membrane; that part of the cytoplasm closed by membranes appeared relatively normal but its density often appeared increased. Next, electron density of the central dramatically increased. Then, membranes proliferated within the cytoplasm in a labyrinth-like manner and an area sequestrated by these membranes enlarged and became more complex structure consisting of vacuoles, electron-dense area and areas of normally-looking cytoplasm connected with convoluted membranes. Finally, a large area of the cytoplasm was transformed into a collection of autophagic vascuoles of different sizes. Virtually identical alternations, albeit with much lower frequency, were seen in terminally ill CJD-affected hansters.

    Streszczenie angielskie: In this study we cloned and analysed partial cDNA of tumor necrosis factor (TNF) and p75 TNF-R receptor of Syrian golden hamster (Mesocricetus auratus). We obtained a 382-bp sequence of TNF and a 148-bp sequence coding for p75 TNF-R. The primers used for the cloning were designed on the basis of inter-species homology, thus presumably can be used for cloning and analysis of TNF and p75 TNF-R genes of other mammals.

    Streszczenie angielskie: The relationship between the secretion of interleukin 10 (IL-10) and nitric oxide (NO) by hamster peritoneal marcophages and their cytotoxic effects on the cells of those two melanoma lines was studied. The nonuniform reaction of macrophages from hamsters bearing two transplantable melanoma lines has been observed. An increase in the cytotoxicity of macrophages from hamsters bearing the amelanotic melanoma line was accmpanied by an inverse correlation between IL-10 and NO secretion. Such a relationship was not found in the case of macrophages from animals bearing the native-melanotic melanoma line. It is suggested that the phenotypical changes of melanomas connected with their progression modified the cytotoxic and secretory activity of the macrophages with regard to IL-10 and NO.
