Tumor necrosis factor (TNF) production and non-Hodgkin lymphoma (NHL) outcome was found to be related to the TNF-308 polymorphism. To explore whether this could be linked to neighboring polymorphisms, we genotyped the TNF-376,-308,-238,-163, lymphotoxin alpha (LTŕ)+252, and HLA DRB1 alleles in 204 patients with NHL and 120 controls. TNF-308A was the only allele associated with higher TNF and its p55 and p75 receptors' levels (P=.009, P=.03, and P=.007) and lower complete remission rates (P=.006). Freedom from progession (FFP) and overall survival (OS) were shorter in patients with TNF-308A (P=.009 and P=.02), null HLA DRB1*02 allele (P=.007 and P.14) or both genetic markers (P=.004 and P.005). Multivariate analysis incorporating international. Prognostic index (IPI) identified TNF-308A (P .0001, relative risk (RR) = 1.63; P .0001, RR=1.51) and null HLA DRB1*02 alleles (P=.015, RR - 1.18; P .0001, RR = 1.25) as independent factors for FFP and OS. These results indicate the existence of least 2 inherited factors involved in NHL outcome. (Blood. 2002; 100 : 3037-3040).

Exposure to grass pollens during the pollen season, reveals in sensitive patients symptoms of allergic rhinitis, conjunctivitis and/or bronchial asthma. It is not well understood why, in some patients, only symptoms of rhinits occur while in others similar exposure causes symptoms of asthma and rhinitis. An association study is reported here, where the possible contribution of human leucocyte antigen (HLA)-DR gene polymorphisms to differential phenotypic expression of symptoms in patients with grass-pollen allergy was determined. HLA-DR genotyping was performed by the polymerase chain reaction with the sequence-specific primers method in 82 patients with symptoms of seasonal allergic rhinitis and/or bronchial asthma and 52 healthy nonatopic control subjects. A significant association was found between HLA-DRB1*02, B5* haploytpe and asthma phenotype in patients with grass-pollen allergy when compared to patients with rhinitis only. Significantly higher total serum immunoglobulin E levels were observed in patients with HLA-DRB1*01 alleles in comparison to patients without these alleles. The data in this study suggest that human leucocyte antigen-DR locus, or other genes in linkage disequilibrium, may play an important role in asthma phenotype expression in patients with grass-pollen allergy as well as in determining total immunoglobulin E levels in these patients.

Background/Aims. The mechanisms of humoral immunological response in chronic hepatitis C are not fully understood. It would be interesting to correlate HLA II alleles with the therapeutic response to interferon alfa2b treatment in patients with chronic hepatitis C. Such correlation has not yet been described. The purpose of the study was to correlate the presence of HLA alleles in chronic hepatitis C patients with results of interferon-alfa2b therapy. Methods. We assessed HCV-RNA presence in serum by RT-PCR and HLA-DR B*1/alleles by PCR-SSP for locus I and II in 54 patients with chronic hepatitis C. All patients were treated with interferon alfa2b for six month. Results of the therapy were evaluated 18 months after the end of treatment. Results. Based on the treatment results (TR - therapeutic response) patients were retrospectively qualified into three groups: sustained responders (SR) - 18.6 p.c. (10/54), relapsers (R) - 48.1 p.c. (26/54), non responders (NR) - 33.3 p.c. (18/54). Allele DRB1* 07 and allele DRB1* 13 were more frequent among patients in NR and TR group, respectively. Observed diffrences were not statistically significant (p 0.05). Conclusion. In the group of 54 adults with chronic hepatitis C no correlation between the presence of HLA-DRB1* alleles and response to interferon alfa2b therapy was found.

One of the genetic factors associated with the development of alopecia areata (AA) is the HLA locus. The study comprised 52 patients with AA aged 10 to 64 years. The frequences of HLA-DRB alleles in the patients and controls were compared. The control group comprised 152 healthy persons. Familia occurrence of AA was seen in 7 (13.5 p.c.) cases. A significantly lower frequency of HLA-DRB1*03 was observed in patients with AA comparison with the control group. In all patients with AA, alleles HLA-DRB1*15/*16 occurredd more frequently than in the control group, but this was not significant after correction. In the group of patients with more severe forms of alopecia areata (alopecia areata totalis/alopecia areata universalis) there was no signficant difference in HLA-DR allele distribution.
