    Streszczenie polskie: Przedstawiono przypadek 39-letniego mężczyzny, przyjętego do Kliniki Urologii Centrum Onkologii z podejrzeniem nowotworu lewego jądra. Wstępna diagnoza histopatologiczna wykonana po orchidectomii wskazywała na nasieniaka (seminoma). Badanie cytogenetyczne na materiale guza wykonano przy użyciu technik klasycznego barwienia różnicowego chromosomów metafazowych (GTG) oraz fluorescencyjnej hybrydyzacji in situ (FIHS). Stwierdzono obecność kariotypu hipotetraploidalnego z występowaniem licznych zmian strukturalnych, dotyczących głównie chromosomów 1, 6, 18, X. Nie stwierdzono aberracji obejmujących krótkie ramię chromosomu 12-12p. Obecność amplifikacji materiału 12p jest specyficzna dla wszystkich guzów jąder (TGCT - Testicular Germ Cell Tumor), niezależnie od typu histologicznego. Komórki badanego nowotworu wykazywały natomiast wysoki stopień multiplikacji materiału pochodzącego z chromosomu 18. Aberracje dotyczące tego chromosomu nie są typowe dla guzów jąder, są natomiast charakterystyczne dla chłoniaków nieziarniczych. Uzyskany wynik poddawał w wątpliwość pierwotną diagnozę. Powtórna analiza preparatów histologicznych, wraz z dodatkowym badaniem immunohistochemicznym i histochemicznym, pozwoliła na ostateczne ustalenie rozpoznania jako chłoniak rozlany z dużych komórek B (DLBCL - Diffuse Large B-Cell Lymphoma).

    Streszczenie angielskie: A 39-year-old patient, edycja suspected of testicular cancer was admitted to the Urology Clinic and left-sided inguinal orchidectomy was performed. The preliminary diagnosis of seminoma testis sinistri, grade I was estabished. Cytogenetic study was performed using subsequent conventional G-band analysis and fluorescent in situ hybridization (FISH) method using specific DNA probes. Analysis of G-banded metaphases revealed a hypotetraploid chromosome complement with many nonrandom chromosome aberrations. Those changes mainly comprised 1, 6, 18 and X chromosomes. Aberrations of 12p material were not detected, especially isochromosome i(12p) was absent. The amplification of the short arm of the chromosome 12, especially as the i(12p) marker chromosome, has been found as a highly non-random chromosome abnormality associated with testicular germ cel tumors (TGCT). Amplification of 12p material is specific for all histological types of such tumors. In 80-90 per cent of cytogeneticall analysed TGCT an overrepresentation of complete short arm of chromosome 12, as the i(12p) marker, has been frequently reported. Furthermore, studies using FISH methods have revealed that 12p aberrations occur in every i(12p)-negative TGCT. In such cases multiplicated 12p material has been found in markers substitutive for i(12p). In the current studied case, analysed cells showed multiplication of normal copy of chromosome 18 and some chromosomal markers, that involved chromosome 18 material.     Streszczenie angielskie: According to the REAL/WHO classification, the diagnosis of mantle cell lymphoma (MCL) should be based on clinical, histopathological, immunological and cytogenetic or molecular data. This study is based on 13 cases, which were initially diagnosed as MCL with the use of conventional cytogenetic method and fluorescent in situ hybridization (FISH). MCL is associated with a specific cytogenetic aberration t(11;14)(q12;q32). The chromosomal analyses confirmed the MCL diagnosis in four cases. A neartetraploid cell line and two copies of t(11;14) were observed in three cases. These results correspond with a blastoid variant of MCL, accompanied by aggressive course and poor prognosis. The presence of karyotype with t(11;14) as the sole anomaly predicts an intermediate clinical outcome. Six patients had normal karyotypes, which is characteristic for the typical form of MCL, associated with a better prognosis. In this study we show that detection of chromosomal abnormalities is useful in diagnosis of MCL and has some prognostic significance.
