    Streszczenie angielskie: The expression of type I collagen has been compared in fibroblast and osteoblast cultures of a patient with moderately severe osteogenesis imperfecta (OI) type IV, with respect to control cells. Electrophoretic analysis of type I collagen showed that both OI osteoblasts and fibroblasts synthesized normal chains with delayed migration. However, the osteoblasts contained a higher proportion of abnormal chains than fibroblasts from the proband. Pulse-chase experiments showed that the trimers containing abnormal chains were cleared more rapidly from osteoblasts than fibroblasts. Moreover, the collagen secreted by OI osteoblests had thermal stability 1řC higher than collagen secreted by OI fibroblasts. These results suggest that the abnormal collagen in osteoblasts may be more resistant to intra- and extracellular degradation and my thus have better survival than in fibroblasts. This finding could have implications for understanding the clinical phenotype of OI.

    Streszczenie angielskie: Breast cancer cell line cultures provide an experimental model in which the action of a variety of mitogens and proapoptotic factors can be studied in vitro. Expression of LHRH and its receptor has been demonstarted in human breast adenocarcinoma, MCF-7 cell line, ER-positive. Antiproliferative effect of LHRH-analogs on breast cancer cells has been revealed. The aim of the present study was to examine the effect of different concentrations (10(-6) and 10(-7) g/ml) and time of exposition (24 hr, 48 hr and 72 hr) of Decapeptyl, an agonistic analog of LHRH, on the expression of wild and mutant p553 and bcl-2 gene products as well as on the proliferative activity (expressed as [3H] thymidine incorporation an dProliferating Cell Nuclear Antigen-PCNA and Mitotic Proteins-MPM2 antigens involvement) in MCF-7. Besides we compared the effects of Decapeptyl with the influence of Tamoxifen alone (10 ćM) and Tamoxifen along with Decapeptyl (10 ćM, 10(-6)g/ml respectively). Decapeptyl, independently of the used dose and time of exposition, did not alterradioactivity detected in studied cells in comparison with the control group. Furthermore, label found after 24 hours of exposition was comaprably high as after Estardiol treatment. Thymidine incorporation after Decapeptyl was consistently higher than afetr Tamoxifen and Tamoxifen/Decapeptyl together. Decapeptyl, in dose and time independent manner, do not change the number of PCNA-immunopositive cells, and only at higher concentration [10(-6) g/ml] decreased immunodetection of MPM2...

    Streszczenie angielskie: A series of carbamate derivatives of Hoechst 33258 was prepared as potential anticancer agents. These new compounds (1-4) were readily prepared in good yields by addition of chloroethyl, bromoethyl, chloropropyl or 4-(chloromethyl)phenyl isocyanates to Hoechst 33258. Their cytotoxic activity was evaluated on human breast cancer MCF-7. Compounds 1-4 were more cytotoxic than Hoechst 33258. In particular derivatives 4, the most active of the series, is up to 3 times more potent than Hoechst 33258. The DNA-binding ability of these compounds was evaluated by an ultrafiltration method using calf thymus DNA. These data show that in broad terms the cytotoxic potency of 1-4 in cultured breast cancer MCF-7 cells incerases, in accord with their increases in DNA affinity, as shown by the binding constant values.

    Streszczenie polskie: Badaniami objęto 30 kobiet leczonych z powodu niepłodności związanej z zespołem policystycznych jajników. Za kryterium diagnostyczne PCO przyjęto: występowanie cykli bezowulacyjncyh i obraz jajników w badaniu ultrasonograficznym. Pacjentki leczono jedną z metod: przy pomocy laparoskopowej elektrokauteryzacji jajników lub przezpochwowej punkcji pęcherzyków. Porównywano ilość cykli owulacyjncyh po zabiegu i kumulacyjny wskaźnik ciąż w przebiegu trzymiesięcznej obserwacji. Skuteczność leczenia obydwiema metodami była zbliżona. Zaletami punkcji pod kontrolą USG wydaje się być łątwość wykonania oraz mniejsze obciążenie dla pacjentki. Metoda ta ponadto daje możliwość hodowli niedojrzałych oocytów w warunkach in vitro inastępowegoich zapłodnienia.

    Streszczenie angielskie: Objectives: 30 patients reated for infertility associated with PCO were analyzed. Anovulatory cycles and US scans of ovaries were taken as criteria for PCO. Mateirals and methods: Patietns were treated either by the laparoscopic electrocauerisation (n = 16) of ovaries or by the ultrasonografic punction of follicles (n = 14). Number of ovulatory cycles, cumulative pregnancy rate and hormonal changes were analyzed. Results: We observed the same efficacy of both methods in terms of ovulatory cycles and pregnancy rates. The cumulative pregnancy rate was 37,5 p.c. in laparoscopic and 35.7 p.c. in ultrasonographic groups during the 3 months' observation. Conclusions: Ultrasonographic punction has appeared to be easier to do and safer for the patient and have the same pregnancy rate. It also gives opportunity for in vitro culture and maturation of obtained oocytes.

    Streszczenie angielskie: Melatonin in the in vitro conditions inhibits cell growth and proliferation of estrogen sensitive (ER+) cell line MCF-7 in culture. In the present study, during a 48-hour incubation melatonin at a concentration of 10-**5M inhibited [3H]thymidine incorporation into cancer cells at the level of 69.52 p.c. ń 10.99 p.c. Melatonin had no inhibitory effect on the physiological stimulatory action of estradiol. Tamoxifen added to the medium modulated the melatonin action only when the latter was added 24 hours after tamoxifen (46.45 p.c. ń 4.40, p 0.05). Tretinoin added to the culture caused a statistically significant reduction in [3H]thymidine incorporation into the cancer cells, compared to he melatonin and tretinoin groups, when treatment with retinoid was synergistic (39.05 p.c. ń 5.44, p 0.05) or sequential (tretinoin and after 24h melatonin) (39.96 p.c. ń 1.55, p 0.05). This was confirmed by immunocytochemical investigations, which showed a statistically significant reduction in the percentage of PCNA- and Ki67-positive cells. Apart from the inhibitory effect on MCF-7 cell proliferation retinoids induce the apoptotic pathway in a dose-dependent manner. Melatonin added to the culture enhances this effect, which may indicate the potential for the use of both substances in the treatment of breast cancer in women.

    Streszczenie angielskie: Cultured skin fibroblasts from a proband with a lethal form of osteogenesis imperfecta produce two forms of type I collagen chains, with normal and delayed electrophoretic migration; collagen of the proband's motehr was normal. Peptide mapping experiments localized the structural defect in the proband to ŕ1(I) CB8 peptide in which residues 123 to 402 are spaned. Direct sequencing of amplified cDNA covering this region revealed a G to A single base change in one allele of the ŕ1(I) chain, that converted glycine 388 to arginine. Restriction enzyme digestion of the RT-PCR product was consistent with a heterozygous COL1A1 mutation. The novel mutation conforms to the linear gradient of clinical severity for the ŕ1(I) chain and results in reduced thermal stability by 3řC and intracellular retention of abnormal molecules.
