    Streszczenie angielskie: The aim of the present study was to examine the influence of 3-month administration of the typical neuroleptic haloperidol (1 mg/kg/day) and the atypical one clozapine (30 mg/kg/day) on the expression of the NMDA-R1 mRNA in different brain structures using in situ hybridization in rats. A long-term treatment with haloperidol decreased the NMDA-R1 mRNA level in intermediate and caudal parts of the caudate-putamen and in more caudally localized regions of parietal and frontal cortices, but increased it in the CA1 region of the hippocampus. No significant changes in the nucleus accumbens, insular cortex, CA3 and dentate gyrus of the hippocampus were found after haloperidol administration. Clozapine did not influence the NMDA-R1 mRNA expression in the hippocampus, as well as in the intermediate and caudal regions of the caudate-putamen, but significantly increased it in the rostral region of the latter structure, in the nucleus accumbens and insular cortex. The present study suggests that both these neuroleptics influence the expression of the mRNA of the NMDA-R1 subunit in brain structures which are thought to be important for development of psychotic symptoms.

    Streszczenie angielskie: Chronic unpredictable stress (CUS) model of depression is one od the well validated animal models of depression. In this paper, we report the results of investigations into dopaminergic D-1 and serotonergic 5-HT-2A receptors in the brain of rats subjected to CUS procedure and treated chronically with imipramine. We have examined the dopaminergic D-1 ([3H-SCH 23390) in the limbic area and serotonergic 5-HT-2A ([3H-ketanserin) receptors in the cerebral cortex by a saturation radioligand binding method in rats subjected to CUS paradigm, imipramine, both CUS and imipramine and control animals. CUS procedure resulted in a significant 36 p.c. increase in the D-1 receptor density in the limbic system, which was attenuated by chronic imipramine treatment. Also a 21 p.c. increase in the density of 5-HT-2A receptors in the cerebral cortex induced by CUS was reduced by chronic imipramine treatment. The present data indicate that the increases in the density of brain D-1 and 5-HT-2A receptors of rats subjected to CUS, which are "normalized" by imipramine, might be involved in the pathophysiology of :animal depression" (and, thus, in pathophysiology of human depression) and in the mechanism of antidepressant tharapy.

    Streszczenie angielskie: The aim of the present study was to find out whether the metabotropic receptor 1 (mGluR1) and group II mGluRs, localized in the striatum, are involved in antiparkinonian-like effects in rats. Haloperidol (1 mg/kg ip) induced parkinsonian-like muscle rigidity measured as an increased resistance of a rat's hind foot to passive flexion and extension at the ankle joint. (RS)-1-aminoindian-1,5-dicarboxylic acid (AIDA; 0.5-15 ćg/0.5 ćl), a potent and selective mGluR1 antagonist, or (2R,4R)-4-aminopyrrolidine-2,4-dicarboxylate (2R,4R-APDC; 7.5-15 ćg/0.5 ćl), a selective group II agonist, was injected bilaterally into the striatum of haloperidol-treated animals. AIDS in doses of 7.5-15 ć/g/0.5 ćl diminished the haloperidol-induced muscle rigidity. In contrast, 2R,4R-APDC injections were ineffective. The present results may suggest that the blockade of striatal mGluR1, but not the stimulation of group II mGluRs, may ameliorate parkinsonian muscle rigidity.

    Streszczenie angielskie: The article reviews presently commonly accepted concepts of neuronal basis of neuroleptic-induced extrapyramidal side effects. The data obtained both, in humans and laboratory animals, point to the blockade of a large number of the striatal dopamine D2 receptors by neuroleptics as a primary cause of these disturbances. This phenomenon leads to the appearance of parkinsonian symptoms shortly after therapy commencement. On the other hand, chronic administration of neuroleptics evokes supersensitivity to dopamine connected with the increased number of D2 receptors and supersensitivity of D1 receptors, which can be significant for the development of tardive dyskinesia. Primary and secondary changes in the function of dopamine receptors lead to partially opposite, pathological changes in the activity of neuronal pathways connecting the basal ganglia. Besides functional changes, neuroleptic-induced lesions of the striatal neurons and genetic predispositions can also play a role in tardive dyskinesia.

    Streszczenie angielskie: Several animal models of "depression" have been examined. One of them is chronic unpredictable stress (CUS)-induced deficit of fighting behavior in rats. In the present study, we compared the effects of two antidepressants (fluoxetine or fluvoxamine) and three anxiolytics (buspirone, lorazepam or oxazepam) on the electric footshock-induced fighting behavior in the pairs of male Wistar rats exposed to CUS procedure (16-day application of various unpredictable stressors). It was found that, in chronically stressed rats, the number of fighting attacks was significantly reduced (by about 70 p.c.). Prolonged (for 14 days) treatment of rats with fluoxetine or fluvoxamine (both at the dose of 10 mg/kg/day) counteracted the deficit of aggression induced by the chronic stress,. On the contrary, the anxiolytics: lorazepam (0.5 mg/kg/day), oxazepam (5 mg/kg/day) or buspirone (0.2 mg/kg/day) administered for 14 days, did not modify the deficit of fighting induced by CUS procedure. It must be underlined that prolonged treatment with all used drugs did not change the intensity of fighting in normal (unstressed) rats. In conclusion, prolonged tratment with antidepressant drugs prevents the CUS-induced deficit of fighting behavior, whereas no beneficial effect of anxiolytic agents was found.

    Streszczenie polskie: Wstęp. Ostatnie badania sugerują, że te same czynniki genetyczne mogą odpowiadać zarówno za rozwój nadciśnienia tętniczego, jak i nefropatii cukrzycowej, ale molekularny mechanizm leżący u podłoża obydwu tych schorzeń nie został jeszcze poznany. Locus Sa zidenttyfikowano jako ulegający wzmożonej ekspresji w nerakch szczurów z nadciśnieniem etniczym. Opisano związek polimorfizmu długości fragmentów restrykcyjnych (Pst1) produktu polimerazowej reakcji łańcuchowej (PCR-RFLP) tego genu z występowaniem nadciśnienia tętniczego u osób bez cukrzycy. Celem pracy było zbadanie związku pomiędzy polimorfizmem Pst1 genu Sa a występowaniem cukrzycowej choroby nerek w stadium mikroalbuminurii lun jawnej nefropatii u chorych na ukrzycę typu 2. Ponadto oceniono zależność między badanym polimorfizmem a obecnością lub brakiem nadciśnienia tętniczego. Materiał i metody. Plimorfizm genu Sa oceniono u 693 chorych na cukrzycę typu 2, u których stan zaawansowania powiklań nerkowych oceniono na podstawie analizy dotychczasowych danych klinicznych oraz pomiarów stosunku wydalania mikroalbumin/kraetyniny w 3 próbkach moczu. W powyższej grupie znalazło się 127 chorych z jawną nefropatią (białkomocz), 323 chorych z mikroalbuminurią oraz 243 chorych, u których stwierdzono normoalbuminurię z czasem trwania cukrzycy powyżej 10 lat stanowiących grupę kontrolną. Z 693 chorych na cukrzycę typu 2 utworzono następnie grupę chorych z nadciśnieniem tętniczym (450) chorych i bez nadciśnienia tętniczego (243 chorych). Wyniki. Nie obserwowano istotnych różnic w rozkładzie genotypów, jak również częstości i występowania alleli polimorfizmu genu Sa Pst1 pomiędzy badanymi grupami...
