    Streszczenie angielskie: We have analysed the heteroplasmy level in 11 individuals from 3 families harbouring the mitochondrial 11778A mutation responsible for Leber hereditary optic neuropathy using last cycle hot PCR. The mutation level exceeded 90 p.c. both in affected and in unaffected individuals. We also checked whether any of the families belonged to the J haplogroup of mitochondrial DNA and obtained a negative results.

    Streszczenie angielskie: Aim of the study: The paper describes the basis of the genetic analysis of the most common mitocondrial mutations in patients with LHON in Poland. Material and methods: The molecular analysis of mitochondrial DNA was conducted in 17 probands and in 5 families with a male descendant with diagnosed or suspected LHON (24 individuals). The four most common mutations responsible for this defect were investigated. Results: The mutations were found in 6 out of 17 probands. Conclusions: Genetic investigations in patients with clinical signs of LHON definitely allow to confirm the diagnosis and evaluate the prognosis. They are essential for genetic counseling.
