Streszczenie angielskie: CT-angiography is a non-invasive method, alternative to cerebral angiography in the diagnosis of intracranial aneurysms. The aim of the study was the assessment of the efficiency of CT-angiography in a large material, which has been mostly verified with angiography ad/or surgery. CT-angiography was performed in 196 patients with intracranial bleeding. Seventy-three (73) patients underwent also cerebral angiography (CA), 121 were operated on, 74 of them solely on the basis of CT-angiography. A spiral CT scanning (2 mm slices, 1-1.5 pitch, 1 mm-gap reconstruction) combined with an injection of 120 mL of contrast medium (5 mL/sec) was used. Subsequently three-dimensional MIP reconstructions were received, supplemented in some cases with SSD, VRT and MPR. CT-angiography revealed 128 aneurysms in 106 patients, located mostly in anterior communicating, middle cerebral and internal carotid arteries. The size of most aneurysms was 6 - 10 mm, but nearly one-third of them did not exceed 5 mm. In the group of patients who underwent CA, there was only 1 false negative CT-angiography result, while in 7 patients CT- angiography allowed for the diagnosis of small aneurysms which were not clearly visible on CA. CT-anioggraphy was superior to CA in assessment of aneurysm morphology by 3 independent radiologists in 17 out of 22 cases. CT-angiography findings were confirmed during surgery in all 74 patients. CT-angiography is highly effective in the diagnosis of intracranial aneurysms and the evaluation of their morphology. In our opinion, in most cases, CT-angiography is precise enough to plan a surgical or endovascular procedure.

Streszczenie angielskie: Rejecting tissue and organ grafts shed cellular debris from damaged cells. Cellular debris contains fragments of nuclei with their genetic material. The question arises what is the fate of donor. DNA in recipient fluids and tissues. Is it enzymatically disintegrated and becomes a waste product or it is utilized by recipient cells. We have shown, using sex-mismatched transplants and the Sry-gene fragment PCR detection method, that at the time of rejection recipient tissue contain donor DNA. The concentration of donor DNA did not paralled the concentration of live donor passenger cells. There were differences in donor DNA concentration depending on whether heart, skin or BMC were transplanted. Moreover, there was more donor DNA in recipient tissues than in control syngeneic transplants. Interestingly, a relatively, a high donor DNA concentration was detected in syngeneic recipients reflecting the extent of non-immune pre-transplantation ischemic damage of the graft.

Streszczenie angielskie: This study indicates that circulating progenitors of bone marrow orgin give to cells with smooth muscle-like properties during formation of neointimal thickenings in the arterial wall after allotransplantation and after balloon injury. A segment of abdominal aorta was transplanted from female F344 to male LEW rats, and the grafts were analyzed for male cells by using the SRY gene a marker. Immunostaining demonstrated that CD45-positive leukocytes made up 35-45 p.c. of the neointimal cells during the 8-week period eamined. Concurrently, up to 70 p.c. of the neointimal cells were of host origin, as shown by real-time polymerase chain reaction for the SRY gene (Y chromosome). This suggests that te neointima contained host cells also of noniflammatory character. Accordingly, many cells positive for smooth-muscle ŕ-actin were detected in this layer. To explore the possible bone marrow origin of allograft cells, female LEW rats were irradiated and substituted with bone marrow from male LEW rats. Subsequently, the animals received an aortic transplant from female F344 rats or were exposed to a balloon injury of the carotid artery. Immunostaining and real-time polymerase chain reaction confirmed the above findings, but the fractions of leukocytes and SRY-positive cells were lower in the carotids than in the allografts. Combined primed in situ labelin and immunostaining verified that not only inflammatory but also smooth muscle-like cells of male origin appeared in the vessel wall in both situations. These observations suggest that the smooth-muscle cells that participate in the development of neointimal lesions during vascular disease may, in part, originate from circulating progenitors.
