Streszczenie angielskie: Structures of five new O-specific polysaccharides of the lipopolysaccharides of bacteria Proteus were established. Four of them, Proteus penneri 4 (072), Proteus vulgaris 63/57 (037), Proteus mirabilis TG 277 (069), and Proteus penneri 20 (017), contain O-acetyl groups in non-stoichiometric quantities, and the polysaccharide of P. penneri 1 st structuraly related to that of P. penneri 4. The structures were elucidated using NMR spectroscopy, including one-dimensional 1H nd 13C NMR, two-dimensional 1H, 1H correlation (COSY, TOCSY), 1H, 13C heteronuclear multiple-quantum coherence (HMQC) and multiple-bond correlation spectroscopy, as well as nuclear Overhauser effect spectroscopy (NOESY or ROESY), along with chemical methods. The structural data obtained are useful as the chemical basis for the creation of the classification scheme for Proteus strains.

Streszczenie angielskie: The O-specific polysaccharide chains (O-antigens) of the lipopolysacchardides of five Proteus strains, P. vulgaris 017, P. mirabilis 016 and 033, and P. penneri 31 and 103, were found to contain phosphate groups that link the non-sugar components, namely, ethanolamine and ribitol. From them, the polysaccharides of P. mirabilis 016 and P. penneri 103 include ribitol phosphate in the main chain and thus resemble ribitol teichoic acids of Gram-positive bacteria. The structures of the polysaccharides were elucidated using NMR spectroscopy, including two-dimensional 1H, 1H correlation spectroscopy (COSY and TOCSY), nuclear Overhauser effect spectroscopy (NOESY or ROESY), and H-detected 1H, 13C, and 1H, 31P heteronuclear multiple-quantum coherence (HMQC), along with chemical methods. The structures determined are unique among the bacterial polysaccharides and, together with data obtained earlier, represent the chemical basic for classification of Proteus strains. Based on structural similarities of the O-specific polysaccharides and serological relationships between the O-antigens, we propose to add P. penneri strain 31 to serogroup 019 and to extend serogroup 017 by including P. penneri strain 16.

Streszczenie angielskie: Gram-negative bacteria of the genus Proteus from the family Enterobacteria-ceae are opportunistic pathogenes which cause mainly wounds and urinary tract infections (UTI), the latter leading to severe complications, such as acute or chronic pyelonephrithis and formation of bladder and kidney stoines. Virulence factors and properties of Proteus sp. mediating infectious process are swarming phenomenon, adherence due to the fimbriae or glycocalyx, flagella, invasiveness, urease, amino acids deaminases, proteases, hemolysins, capsular polysaccharide (CPS), and lipopolysaccharide (LPS. LPS is an integral component of cell wall of bacteria. It also represents the endotoxin which, after being released from bacterial cells, causes a broad spectrum of pathologuical effects leading in severe cases to the septic shock. Lipopolysaccharide consists of three parts: O-specific chain (O-antigen), core and lipid A; all of them have been studied in Proteus LPS. It has been documented that Proteus is a antibenically heterogenous genus, principally because of structural differences in its O-specific polysaccharide chain of LPS. The serological classification of P. mirabilis and P. vulgaris shares 60 serogroups: 22 described for P. vulgaris, 33 characteristic for P. mirabilis and 5 common for both P. mirabilis and P. vulgaris. Serological classification of Proteus penneri still remains to be completed. Proteus O-antigens are branched or linear polysaccharides, built up of oligosaccharide repeating units, varying from a trisaccharide to a hexasaccharide. Acidic O-specific polysaccharides...
