    Streszczenie angielskie: Heat shock protein 27 (Hsp27) belongs to the group of proteins called molecular chaperones protecting normal and tumour cells against many stressors such as hyperthermia, several commonly used chemotherapeutics as well as other apoptotic stimuli. Our study was designed to determine whether heat shock and drugs like cisplatin, etoposide and quercetin have an effect on the expression of heat shock protein 27 in tumour cells such as: HeLa (cervical cancer), Hep-2 (larynx cancer), A549 (lung cancer) and also in normal human skin fibroblasts (HSF) cultured in two-dimensional (2D) and three-dimensional (3D) conditions. Our results indicate that Hsp27 expression is drug- and cell-type specific and depends on the culture model as well. HeLa, Hep-2 and HSF cell cultured in 3D model appeared to be more resistant to stimulatory or inhibitory effects of the applied drugs than cells cultured in 2D model. Only A549 cells cultured in 3D culture model appeared to be more susceptible and reacted to drugs and heat treatment with increased Hsp27 expression.

    Streszczenie angielskie: The present study was designed to investigate whether quercetin, a very common flavonoid widely distributed in many plants, can induce apoptosis in monkey kidney cells (GMK). Involvement of the expression of heat shock proteins Hsp72, Hsp3, Hsp27 and the significance of cell culture model in this process were also examined. The studies have shown that quercetin alone and in combination with the heat shock can induce apoptosis and necrosis in vitro in the sutided cells, but the percentagae of affected cells did not exceed 3.9 p.c. In the same experimental conditions, the expression of Hsp73, Hsp72 and Hsp27 increased in cells cultured in two-dimensional systm and decreased in three-dimensional model. This indicates that strong inhibition of heat shock proteins in GMK cells is not correlated with an adequate increase in the sensitivity of cells to undergo apoptosis. It also shows that the sensitivity of cells, manifested by Hsp expression and apoptosis induction, depends on the culture model and culture conditions.

    Streszczenie angielskie: The present study was designed to investigate the correlation between the expression level of Hsp27 and Hsp72 and induction of apoptosis in HeLa cells in response to quercetin treatment. Treatment of HeLa cells with quercetin or with 1 hr period of hyperthermia (42ř) increased the number of apoptotic cells. Inhibition of the expression of Hsp72 and Hsp27in tumour cells by anti-sense oligonucleotides, enhanced the induction of apoptosis by quercetin. Heat shock itself had little effect on apoptotic cell death in these cells, but when combinedwith quercetin treatment, caused a significant increase in the number of apoptotic cells. These results suggest that the reduction of Hsps expression in the HeLa cell line promotes the induction of apoptosis by quercetin.

Our study was designed to determine whether heat shock and drugs like cisplatin, etoposide and quercetin influence the expression of heat shock protein 72 in tumour cells: HeLa (cervical cancer), Hep-2 (larynx cancer), A549 (lung cancer) and normal human skin fibroblasts (HSF). Our results indicated that Hsp72 expression was drug and cell-type specific. Cisplatin and etoposide did not induce Hsp72 expression in any tumour cells but cisplatin alone induced Hsp72 expression in normal human fibroblasts. Quercetin inhibited heat shock protein expression in Hep-2 cells but induced in HSF. In thermally shocked tumour cells, the expression of Hsp72 was not altered by cisplatin and etoposide, but in normal HSF cells enhanced expression in normal and tumour cells.
