    Streszczenie angielskie: There are 9 channels of the CIC family in mammals and new others in fishes, plants, yeast and bacteria. The CIC channels are present in different tissues and play a role in transmembrane potential stabilization, transepithelial transport, cell volume regulation, acidification of intracellular organelles. The genetic defects of CIC-1 chloride channel lead to myotonias, the defect in CIC-5 channel to the formation of stones in kidney, while the defect in CIC-Kb channel leads to the Bartter's syndrome.

    Streszczenie angielskie: The glycine receptor belongs to the ligand-gated ion channel superfamily. It is a chloride conducting channel composed of four transmembrane domains. It was previously shown that the second transmembrane domain (M2) of the glycine receptor forms an ion conduction pathway throught lipid bilayers. The amino-acid sequence of the transmembrane segment M2 of the glycine receptor has a high homology to all receptors of the ligand-gated ion channel superfamily. In our report, we have used a synthetic M2 peptide. It was incorporated into a planar membrane of known lipid composition and currents induced by M2 were measured by the Black Lipid Membrane technique. When the planar lipid bilayer was composed of 75 p.c. phosphatidylethanolamine and 25 p.c. phosphatidylserine, the reversal potential measured in a 150/600 mM KCl (sic/trans) gradient was - 19 mV suggesting that the examined pore was preferential to anions, PK/PCl = 0.25. In contrast, when 75 p.c. phosphatidylserine and 25 p.c. phosphatidylethanolamine was used, the reversal potential was +20 mV and the pore was preferential to cations, PK/PCCl = 4.36. SIngle-channel currents were recorded with two predominant amplitudes corresponding to the main-conductance and sub-conductance states. Both conductance states (about 12 pS and 30 pS) were measured in a symmetric solution of 50 mM KCl. The observed single-channel properties suggest that the selectivity and conductance of the pore formed by the M2 peptide of the glycine receptor depend on the lipid composition of the planar bilayer.

    Streszczenie polskie: Magnez jest jednym z najważniejszych wewnątrzkomórkowych kationów w organizmie. Wiele ludzi stosuje preparaty farmaceutyczne zawierające sole magnezu osobno lub razem z innymi mikro- lub makroelementami. Wiadomo, że etanol może wywołać stres oksydacyjny zwłaszcza w hodowlach szczurzych hepatocytów, a także, iż metabolizm etanolu może wywołać peroksydację lipidow. Końcowym produktem peroksydacji lipidów jest między innymi dialdehyd malonowy (MDA). Powszechnie uważa się, że ilość powstającego MDA jest miarą nasilenia procesu peroksydacji. Cel. Celem naszych badań było ocena wpływu dużych dawek jonów magnezu i/lub etanolu na proces peroksydacji lipidów w izolowanych szczurzych hepatocytach. Metodyka. Hepatocyty izolowano z wątrób 3-miesięcznych szczurów rasy Wistar, zgodnie z metodą opisaną przez Seglena. Do badań wykorzystano komórki o żywotności większej niż 95 proc. (oznaczanej błękitem trypanu). Hepatoyty inkubowano w podłożu "Hepatocyte Medium" zawierającym MgCl2 o stężeniu 2,0 i/lub 4,0 mmol/dm3 oraz etanol o stężeniu 150 mmol/dm3, a także mieszaninę obydwu związków. W hepatocytach po 1, 3, 5 godzinach inkubacji oznaczono zawartość dialdehydu malonowego (MDA) zestawem testowym BIOXYTECH**R-LPO-586TM (Oxis International, Inc.). Wyniki. Stwierdzono nasilenie procesu peroksydacji lipidów w hepatocytach inkubowanych z etanolem oraz z jonami magnezu o wyższym stężeniu.     Streszczenie angielskie: Introduction. Magnesium is one of the most important intracellular cations in an organism. Many people use pharmaceutical preparations containing magnesium salts either alone or together with other micro- and macroelements. It is known that ethanol may induce oxidative stress in rat hepatocyte cultures and ethanol metabolism can also induce lipid peroxidation. One final product of lipid peroxidation is malondialdehyde (MDA). An increase in the volume of MDA is generaly considered to be an indicator of lipid peroxidation. Aim. The aim of our study was to determine the influence of high doses of magnesium ions and/or ethanol on the process of lipid peroxidation in isplated rat's hepatocytes. Methods. The hepatocytes were isolated from the livers of 3-month old Wistar rats, using Seglen's method. Cells were used if viability was greater than 95 p.c. (assessed) by Trypan Blue exclusion). The hepatocytes were incubated in Hepatocyte Medium containing: 2.0 and/or 4.0-mol/lMgCl2, 150-mmol/l ethanol, and a mixture of both compounds. After 1, 3 and 5 hours incubation, the concentration of malondialdehyde (MDA) in the hepatocytes was determined by BIOXYTECH**R-LPO-586TM (Oxis International, Inc) assay. Results.
