    Streszczenie polskie: Każdego roku wzrasta zachorowalność na astmę, zwłąszcza wśród dzieci. Pośród wielu przyczyn tego zjawiska zwraca się uwagę na dużą ekspozycję na alergeny domowe, gdyż coraz więcej czasu spędzamy w pomieszczeniach zamkniętych. Wiele doniesień podkreśla znaczący udział alergenów karalucha w tej grupie. Analizie poddano próbki kurzu podchodzące z 39 mieszkań z regiony łódzkiego, pacjentów Poradni Alergologicznej w Zgierzu. W każdym mieszkaniu zebrano jedną próbkę kurzu z kuchni i jedną z sypialni. Stężenie alergenów karalucha w kurzu oznaczano za pomocą gotowych zestawów firmy Indoor Biotechnologies (Wielka Brytania), metodą immunoenzymatyczną ELISA. W 78 próbkach kurzu pobranych z 39 badanych mieszkań stwierdzono występowanie antygenu karalucha Bla g 2 w 43 (55,133 proc.) próbkach, w tym w 62,8 proc. próbkach pobranych z kuchni i 37,2 proc. próbkach z sypialni. We wszystkich 30 mieszkaniach z wykrytym w kurzu antygenem karalucha stężenie antygnu Bla g2 przekaraczało 8 U/g (przynajmniej w jednym z pomieszczeń). W badanej grupie większe stężenia antygenu występowały w starych domach, bez centralnego ogrzewania i w domach o niższym statusie ekonomicznym. Wyniki przeprowadzonych badań wskazują, że częstość występowania antygenu karalucha Bla g 2 w kurzu w domach łódzkich jest znacznie większa od wartości stwierdzanych w innych krajach, a stężenia alergenu Bla g 2 wykazują zależność od charakterystyki badanych mieszkań.

    Streszczenie angielskie: Asthma morbidity increase every year, especially among children. The cause of this is multifactorial, but exposure to high levels of indoor allergens is thought to be one important factor, since we spend the large fraction of time indoors. Next to other popular indoor allergens, many studies underline the importance of cockroach allergen. In this study dust samples from 39 homes were collected. Patients were recruited from Allergy Clinica in Zgierz. In each home the sample from kitchen and bedroom were taken. After extraction Bla g 2 concnetration was measured by ELISA assay (Indoor Biotechnologies, Great Britain). Bla g 2 antigen was detected in 43 (55.13 p.c.) out of 78 samples, in 62.8 p.c. kitchen samples and 37.2 p.c. bedroom samples. In all homes with detected cockroach antigen, the levels were higher then sensitization value 8 U/g (at least in one room). The highest levels of Bla g 2 were found in old houses, without central heating and in houses with lowere income. Our results showed that concentrations of Bla g 2 antigen in homes in Łódź region, are higher than previously reported in other countries and is strongly related to the house characteristics.

    Streszczenie angielskie: Previously, a large two-part inter-laboratory round robin endotoxin assay study was completed. This first study showed that when cotton dust samples, which are practically identical, are assayed for endotoxin that the intra-laboratory results had a very small variation while intra-laboratory results of the sample had a very high variation. In the first part of the study, each laboratory results of the sample had a very high variation. In the first part of the study, each laboratory followed its own in-house assay protocol; but in the second part of the study, when the extraction protocol was standardized, the inter-laboratory results showed a lower variation, which suggested that with further standardization, further reduction of differences between laboratories might be achieved in order that results between laboratories would become more comparable. The results stimulated interest in extending the study to include cotton dust with two levels of endotoxin, standardization of the extraction proteocol, and using the same assay kit from the same production lot. The results of this second round robin endotoxin assay study indicate that differences between laboratories are still high, but most of the laboratories could discern the cotton dusts with the different levels of endotoxin.

    Streszczenie angielskie: Bacgriund: Asthma morbidity increases every year, especially among children, and exposure to high levels of indoor allergens is a very important factor. We evaluated the prevalence and exposure to cockroach (CR) allergen in asthmatic children in Poland, and also tested the hypothesis that asthma with allergy to CR is more severe than with allergy to other antigens. Methods: One hundred and sixty children with asthma were examined, had skin prick tests (SPT) with common and CR allergens, underwent spirometry, and provocation tests to histamine. Children with positive SPT to CR had measured specific IgE levels to this antigen and Bla g 2 concentrations were measured in their homes. Results: The most common allergen, was dust mite 51.3 p.c., followed by pollen 48.8 p.c. and CR allergen 24.3 p.c. In children with CR sensitivity, 13 p.c. had mild ashma, 26 p.c. moderate and 61 p.c. had severe asthma. Their levels forced expiratory volume in one second (FEV1), and te provocative concentration of histamine that caused a 20 p.c. fall in FEV1 (PC20), were statistically lower than in the group of children with other than CR allergies. Bla g 2 antigen was detected in 55.13 p.c. samples. The highest levels of Bla g 2 were found in old houses, without central heating, and in houses with lower income. Conclusion: In Polish children, CR allergen is a very important factor of sensitivity. Concentratins of Bla g 2 in homes are higher than previously reported in other European countries, and are strongly related to the houses' characteristics. Also, children with CR hypersensitivity have severe asthma more often than children with other allergies.
