Streszczenie angielskie: A selective (according to mammalian criteria) histamine (HA) H2-receptor radioligand ([3H]TIOT) was used to characterize HA receptors in duck cerebral cortex by an in vitro binding technique. The specific binding of [3H]TIOT to duck cerebral cortical membranes was found to be rapid, stable, saturable, reversible, and of high affinity. Saturation analysis resulted in a linear Scatchard plot suggesting binding to a single class of receptor binding sites with high affinity (Kd = 19.5 nM) and high capacity (Bmax - 356 fmol/mg protein). Competition studies showed the following relative rank order of potency of various HA receptor ligands to inhibit the [3H]TIOT binding: antagonists, tiotidine rantidine izolantidine ň cimetidine mepyramine thioperamide; agonists, HA ň 4-methylHA 2-methylHA dimaprit Rŕ-methylIHA. The biphasic nature of the displacement curve for HA and the effect of 5'-guanylimidodiphosphate indicate with [3H]TIOT represent either avian-specific H2-like HA receptors or a novel subtype of HA receptors, coupled to a signalling athway other than the adenylyl cyclase/cyclic adenosine monophosphate one.

Streszczenie angielskie: It has been found that trypsin and ŕ-chymotrypsin inhibitors isolated from Ascaris suum act embryotoxically and teratogenically on White Leghorn chicken embryos. Mortality rate for the chicken embryos on day 15 of incubation was 45.0 ń 3.5 p.c. after injection of trypsin inhibitor and 44.0 ń 3.5 p.c. after adminstration ŕ-chymotrypsin inhibitor. Gross examination of surviving embryos and their dissection revealed pathological changes (abdominal dropsy, umbilical hernia, subcutaneous oedema, hemoperitoneum, hemopericardium), symptoms indicating retardation in growth (lack of down, retarded ossification of long bones, decreased mean body weight) as well as malformations (schistocelia, micrognathia, cyclopia, crossed beak, cranial deformities) after injection of inhibitors from Ascaris. The highest incidence of embryos with pathological changes and malformationsd was foundf after administration of ŕ-chymotrypsin inhibitor. The most commonly occurring abnormality was schistocelia (21.4 ń 3.88 p.c.). Growth malformations were not found in the control groups. The trypsin and ŕ-chymotrypsin inhibitors present in Ascaris homogenate have a significant disturbing effect on ther development of the chicken embryo.

Streszczenie angielskie: This study has demonstrated that the short and long form of the pituitary adenylate cyclase-activating polypeptide (PACAP), i.e. PACAP27 and PACAP38, moderately but significantly, and in a concenration (0.5-5 ćM)-dependent manner, stimulated inositol phosphates (IPs) accumulation in myo-[3H]inositol-prelabeled cerebral cortical and hypothalamal slices of chick and duck, and in slices of rat cerebral cortex; both peptides had no effect on IPs formation in rat hypothalamus. Vasoactive intestinal peptide (VIP; 0.5-5 ćM) weakly enhanced IPs accumulation in chick hypothalamus, had no significant action in chick cerebral cortex (in fact there was a tendency to attenuate the IPs response in this tissue), and slighty, but significantly, inhibite the IPs accumulation in rat cerebral cortex. VIP showed no activity in rat hypothalamus. It is cincluded that tha stimulatory action of PACAP on phosphoinositide metabolism in avian cerebral cortex, similar to rat cerebral cortex, is mediated via phospholipase C-linked PAC1 type receptors. In chick hypothalamus, however, there may be a component of VPAC type receptors stimulating IPs formation.

Streszczenie angielskie: The chick pineal gland and retina synthesize melatonin in a circadian rhythm with high levels during the night. The rhythmic changes in the hormone production result predominantly from the fluctuation in the activity of serotonin N-acetyltransferase (AA-NAT), a penultimate and key regulatory enzyme in melatonin biosynthesis. The aim of this study was to analyze the effects of an acute and prolonged in vivo treatment with a glucocorticoid dexamethasone (4 mg/kg, ip) on the nocturnal increase in AA-NAT activity in chick pineal gland and retina. In acute experiments, dexamethasone (single dose)-injected chicks were killed after 2 h, while in prolonged experiments the glucocorticoid was given once daily for 7 days and the animals were killed 26-32 h after the last injection. Acute administration of dexamethasone did not affect AA-NAT activity in the chick pineal gland and retina. In the pineal glands and retinas of chicks that were treated with dexamethasone for one week and then killed at the end of the light phase of the 12:12 h light-dark cycle, AA-NAT activity was significantly higher than the enzyme activity found in tissues isolated from the vehicle-treated (control) animals. In addition to that, the nocturnal increase in pineal and, to a lower extent, retinal AA-NAT activity was significantly lower in dexamethasonetreated birds when compared with the respective control groups. It is suggested that prolonged treatment of animals with dexamethasone reduces the amplitude of the rhythmic melatonin production, a phenomenon which may affect chronobiological processes being under control of this hormone.

Streszczenie angielskie: The authors present results of serological examination in 275 pheasants (Phaisanus colchicus) and 273 pigeons (Columba livia f. domestica) for the presence of Chlamydophila (Ch) psittaci IgG antibodies. Using micromethod of complement fixation (CF) test with genus-specific antigen Ch. psittaci (Bioveta, Ivanovice na Han‚, Czech Republic), the seropositivity in pheasants oscillated between 31.5 - 40.4 p.c. No clinical signs of chlamydiosis were detected in pheasants under study. The seropositivity in pigeons ranged between 33.1 - 85.1 p.c. Total 77.1 p.c. positivity with maximal 1: 1024 antibody titre was found in 83 pigeons caught in April 2000, while, in June 2000 positivity was 41.0 p.c. with maximum titre : 512. Similarly, in the year 2001 the seropositivity in the group of 74 pigeons trapped in April reached up to 85.1 p.c. with the highest titre 1 : 1024 and in the pigeons trapped in June positivity decreased to 33.3 p.c. with the titer 1 : 1256. These results prove an acute form of chlamydiosois and suggest that pigeons in spring time are an especially significant source of chlamydiosis for the human and animal population.

Streszczenie angielskie: Transfer of radioruthenium (Ru-106) into hen eggs was examined. Lying hens were administered daily with the radionuclide in drinking water for six days. During the entire study eggs were collected and radioactivity in their homogenised constituents was measured. At the beginning of the experimental period radioruthenium was slightly faster transferred into the albumen but later higher Ru-106 concentrations were recorded in the yolk. It is in contrast to our earlier observations that radiocaesium demonstrates stronger affinity for egg albumen.

Streszczenie angielskie: Pituitary adenylate cyclase-activating polypeptide (PACAP38) potently stimulates cyclic AMP formation in slices of chick cerebral cortex. One- to fifteen-minute pretreatment of slices with 30 nM PACAP38 led to a time-dependent attenuation (when compared with values observed in the control tissue) of the cyclic AMP response produced by subsequent re-stimulation with 1 ćM PACAP38. Concentration-response curve for restimulation with PACAP38 applied at 0.01 - 1 ćM to tissue slices preincubated for 15 min with 30 nM PACAP38 revealed dose-dependent decreases in subsequent cyclic AMP responses by 16-37 p.c. It is concluded that in chick cerebral cortex, the receptors mediating PACAP-driven cyclic AMP responses (PACAP1 receptors) undergo rapid homologous desensitization.
